摘要
目的 :探讨α 生育酚对精索静脉曲张 (VC)大鼠睾丸生殖细胞损害的保护作用。方法 :采用成年雄性Wistar大鼠复制精索静脉曲张模型 ;治疗组 (TG)于手术后 4周开始im ,qdα 生育酚 0 .5mg/(10 0 g体重 )共 4周 ,手术对照组 (CG)动物模型不用药物治疗 ,假手术对照组 (SOG)只行左肾静脉分离 ,不予结扎 ;3组大鼠同等条件饲养 8周。 8周后用原位末端脱氧核苷酸转移酶标记法 (TUNEL)检测睾丸生殖细胞凋亡指数 (AI)。结果 :TG动物左侧睾丸重量显著高于CG(P <0 .0 5 ) ,低于SOG(P <0 .0 5 ) ;TG细胞AI[左侧 (5 .34± 1.2 8) % ,右侧 (5 .87± 1.2 5 ) % ]高于SOG[左侧 (4.72± 1.0 2 ) %、右侧 (4.88± 1.2 8) % ](P <0 .0 5 ) ,而显著低于手术对照组 [左侧 (9.0 5± 2 .5 1) % ,右侧 (7.2 2± 1.35 ) % ](P <0 .0 5 )。结论 :α 生育酚能降低精索静脉曲张大鼠生殖细胞的凋亡指数 ,延缓睾丸细胞损害的病程 。
Objective: To study the effect of antoxidation of α tocopherol on spermategenic cellular apoptosis in experimental varicocele rats.Method: Thirty adult male Wistar rats were randomly divided into 3 groups( n =10), in the treated group (TG) α tocopherol ( 0.5 mg/100 g body weight, one time daily for 4 weeks) was injected into muscle after being created into varicocele model by partial ligation of the left renal vein for 4 weeks, operation controled group (CG)and sham operation group (SOG) were created. All the rats were breed in the similar condition for eight weeks.The apoptosis cells were detected by in situ terminal deoxynucleotityl transferase mediated dTUP nick end lableling(TUNEL).Result: The left venae spermatica of TG and CG were markedly cirsoid compared with SOG( P < 0.001 ), the weight of left testicle in TG was intervened between the CG and SOG ( P < 0.05 ). The changes of histology were extra marked in TG and CG. The apoptosis index of TG was lower than that of CG( P < 0.05 ), and higher than that of SOG( P < 0.05 ).Conclusion: It shows that α tocopherol can decrease the apoptosis index in germ cells of varicocle rats, and slow down the proceeding of testicular injured. There is a certain protecting effect of α tocopherol on the impair induced by experimental varicocele.
