毛细管电泳法测定复方降压片中5组分含量 被引量:4

摘要 目的 建立复方降压片(二方)的高效毛细管电泳5组分同时定量方法。方法 用高效毛细管电泳内标对比法,采用石英毛细管(65 cm× 75 μm,I.D.)有效长度50 cm,50 mmol·L-1硼砂溶液为背景电解质,重力进样10 s(高度7 cm),检测温糜(23±1)℃,紫外检测波长246 nm。结果 测定复方降压片(二方)中维生素B1、磷酸氯喹、氢氯噻嗪、维生素B6和芦丁5组分的相对峰面积的RSD≤4.1%,回收率为94.6%~105.3%,标示百分含量均在96.1%~106.4%。结论 该法可同时分离测定复方降压片中5个组分。 OBJECTIVE To develop a capillary electrophoresis (CE) method for simultaneous determination of 5 components in the Fufangjiangya tablet. METHODS The internal standard contrast method of CE was applied, in which fused silica capillary (65 cm × 75 μm I. D. ) with detection window at 50 cm from anode was employed and 50 mmol · L-1 borax solution served as the running buffer. Other conditions were as follows: sensitivity 0. 010 AUFS, applied voltage 12 kV, hydrostatic injection 10 s (height 7 cm) , assay temperature (23±1)℃ , and detection wavelength 246 nm. RESULTS The RSDs of relative area for the 5 components were all less than 4.1% and their recoveries varied from 94. 6% to 105. 3%. The labling percent contents of the 5 components in the tablet were between 96. 1% and 106. 4%. Both the reproducibility and accuracy of the method met with the technical need of CE. CONCLUSIONS The method is simple, quick and accurate. It can be used to simultaneously assay the 5 components in Fufangjiangya tablet.
