摘要
目的 探讨白藜芦醇苷(PD)对内毒素休克大鼠肺组织中磷脂酶A2(PLA2)的影响。方法 将32只大鼠随机分为4组:单纯手术组(A)、内毒素休克组(B)、PD治疗组(C)和PD预防组(D)。采用酸式滴定法检测肺组织中PLA2活性,RT-PCR法检测sPLA2-ⅡA的基因表达。结果 PD可抑制内毒素所致的PLA2活性增高及sPLA2-ⅡA mRNA表达增强,减轻肺损伤的程度,并且预防性应用的效果优于治疗效果。结论 PD可通过抑制PLA2的活性增高和基因表达增强,从而对内毒素休克大鼠的肺脏具有保护作用。PD可能是一种PLA2抑制剂。
OBJECTIVE: To investigate the effects of polydatin on phospholipase A 2 in rat lung with endotoxic shock. METHODS: Using a rat endotoxic shock model, animals were randomly divided into 4 groups as follows: sham operation group(A), endotoxic shock group (B), polydatin treatment group(C) and polydatin prevention group(D). MAP (mean artery pressure) was measured once half an hour. Lung tissue was collected after the experiment was finished. The activity of PLA2 was measured by acid titration. The gene expression of secretory phospholipase A2 type II A (sPLA2 - II A) was detected by RT-PCR. Furthermore, the histologic damages of lung among groups were investigated by microscope. RESULTS: The activity of PLA2 and the gene expression of sPLA2 - II A were increased by endotoxin, but polydatin restrained these effects of endotoxin. Enough morphological evidence was found in pathological sections of lung. Especially, the protective effects were more obvious in polydatin prevention group. CONCLUSION: Polydatin was found to have prophylactic and therapeutic effects on acute lung injury following endotoxic shock and the prophylactic effect tends to be better than the therapeutic effects. Polydatin may be a phospholipase A2 inhibitor.
