摘要
阳离子染料灿烂甲酚蓝(BCB)在适宜浓度的阴离子表面活性剂十二烷基硫酸钠(SDS)或十二烷基磺酸钠(SLS)存在时形成二聚体。研究了以此形成的现场二聚体作为荧光探针测定蛋白质的可能性。结果表明,二聚体具有弱荧光,其荧光强度的回升与体系中蛋白质的量呈线性关系,线性范围为0~7.8μg·ml-1,检出限为3.89×10-3μg·ml-1。用于人血清白蛋白的测定,结果满意。
It was found that in the presence of appropriate amounts of anionic surfactant of sodium lauryl sulfate (SDS) or sodium lauryl sulfonate (SLS), the cationic dye brilliant cresol blue (BCB), is dimerized in the respective solution. And in the further study of the fluorescence behavior of BCB dimer, it was found that the weak fluorescence intensity of the dimer is enhanced by its reaction with human serum albumin and a linear relationship between the fluorescence intensities and the serum albumin concentrations is found in the range of 0~7.8μg·ml^(-1), and the detection limit found is 3.89×10^(-3)μg·ml^(-1). Based on these facts, the BCB dimer in solution containing appropriate amounts of SDS or SLS is used as fluorescence probe for the determination of protein and satisfactory results are obtained.
