摘要
以吖啶橙在十二烷基硫酸钠介质中生成的二聚体作为荧光探针,讨论了其与牛血清白蛋白相互作用的情况。实验表明,蛋白质的加入可使低荧光的吖啶橙二聚体荧光恢复,且其荧光增强的程度与体系中蛋白质的加入量在一定浓度范围内呈线性关系。在优化实验条件下,线性范围为0.8—34mg/L,检出限为0.126mg/L。
In the presence of appropriate amounts of anionic surfactant of sodium dodecyl sulfate(SDS),the cationic dye acridine orange(AO)is dimerized.It was found that the weak fluorescence intensity of the AO dimer is enhanced by its reaction with bovine serum albumin(BSA)and a linear relationship between the fluorescence intensities and the serum albumin concentrations.Based on these facts,the AO dimer in solution containing appropriate amounts of SDS is used as fluorescence probe for the determination of protein.The linear range for the determination of BSA was 0.8—34mg/L.This method shows a detection limit of 0.126mg/L.
