卵巢上皮性肿瘤组织中ERK的表达与活化

摘要 目的 探讨上皮性卵巢肿瘤组织中细胞外信号调节激酶 (ERK)的表达与活性及其与卵巢上皮性肿瘤临床病理因素的关系。方法 应用免疫组织化学技术 ,检测 4 9例卵巢上皮癌组织 ,2 6例良性卵巢上皮性肿瘤组织及 10例正常卵巢组织中ERK1/2蛋白和活性ERK1/2蛋白表达。结果 ERK1/2在卵巢上皮性癌组织中的表达阳性率为6 3% ,显著高于正常卵巢组织 (阴性 )和卵巢上皮性肿瘤组织 ( 2 7% ) (P <0 .0 1)。Ⅲ~Ⅳ期卵巢癌组织中的ERK1/2蛋白表达水平高于Ⅰ~Ⅱ期卵巢癌 (P <0 .0 5 )。活性ERK1/2在卵巢上皮性癌中的阳性表达率为 6 5 % ,明显高于卵巢良性肿瘤组织中活性ERK1/2的阳性表达率 ( 19% ,P <0 .0 1)。手术病理分期Ⅲ~Ⅳ期卵巢癌组织的活性ERK1/2表达阳性率为 80 % ,Ⅰ~Ⅱ期为 13例中 3例 ,两者比较 ,差异有显著性 (P <0 .0 1)。结论 ERK蛋白的过表达在卵巢上皮性癌患者的发生、发展中可能起重要促进作用 。 Objective To investigate the expression and activation of extracellular signal regulated kinase (ERK) and its significance of epithelial ovarian tumor.Methods Expression of ERK protein and active ERK was examined by immunohistochemical method in the epithelial ovarian tumor of 49 patients,benign epithelial ovarian tumor of 26 patients and normal ovarian tissue of 10 patients.Results The positive percentage of ERK1/2 protein in epithelial ovarinoma was 63%,which was significantly higher than that in benign tumors(27%) and normal ovarian tissues (negative)(P<0.01).The positive percentage of ERK1/2 protein in stage Ⅲ~Ⅳ epithelial ovarian cancer was significantly higher than in stage Ⅰ~Ⅱ epithelial ovarian cancer(P<0.05).The positive percentage of active ERK1/2 in epithelial ovarian carcinoma was 65%,which was obviously higher than that in benign tumors(19%,P<0.01).The positive percentage of active ERK1/2 in stage Ⅲ~Ⅳ of epithelial ovarian cancer was 80%,while there was 3 positive cases in stage Ⅰ~Ⅱ out of 13 cases,there was significantly difference between them(P<0.01).Conclusion The overexpression and activation of ERK may play an important role in the initiation and development of human ovarian cancer,which is a prognostic indicator in patients with epithelial ovarian cancer.
