Antimicrobial and Preservative Effects of Codonopsis Pilosula Extract and Identification of its Active Ingredients

Codonopsis pilosula(C.pilosula),Astragalus membranaceus(A.membranaceus)and Angelica sinensis(A.sinensis)are three common medicinal and dietary tonifying herbs.In this research,the antimicrobial effects of their aqueous and ethanol extracts were analyzed by using the filter paper diffusion method and microdilution method.The results showed that the aqueous extracts of these three medicinal herbs had no significant inhibitory effect on the growth of four common food-contaminated microorganisms:Escherichia coli(E.coli),Staphylococcus aureus(S.aureus),Candida albicans(C.albicans)and Saccharomyces cerevisiae(S.cerevisiae).Only the C.pilosula ethanol extract(denoted as CEE)could significantly inhibit the growth of S.aureus.The antimicrobial and preservative effects of CEE were investigated by using carrot juice as a model.It was found that CEE exhibited significant synergistic antimicrobial and preservative effects with the chemical preservative benzoic acid.When benzoic acid was reduced to half of its conventional food preservative dosage,the combined addition could completely inhibit the growth of S.aureus within 6 d.CEE was further analyzed by high-performance liquid chromatography(HPLC)and primary mass spectrometry(MS1),and lobetyolin was preliminarily identified as the main active ingredient.The minimum inhibitory concentration(MIC)of lobetyolin was determined to be 40μg/mL when used alone.The cell counting kit-8(CCK-8)assay showed that both CEE and lobetyolin exhibited low cytotoxicity on human normal liver QSG-7701 cells at the preservative dosage,suggesting promising potential for developing safe and effective food preservatives from traditional Chinese medicinal herbs. 展开更多

Ecological Factors Drive the Accumulation of Active Components in Codonopsis pilosula

Codonopsis pilosula is a major Qi-tonifying medicinal herb,and its active composition is analyzed systematically.However,the relationship between its production origins and commodity specification grades with the active composition of C.pilosula lacks systematic research.This study integrates the HPLC and UV-Vis methodologies to evaluate the quality of C.pilosula from commodity specification grades and different origins,and it explores the correlation between ecological factors and production origins with active components.Here,network pharmacology is used to determine that lobetyolin,syringin,and tangshenoside I have potential efficacy in treating pulmonary fibrosis and oxidative stress.The HPLC and UV-Vis methods were employed to quantitatively analyse the levels of five active compounds from different origins and commodity specification grades.Ecological factors were collected from the different production origins with ArcGIS,and correlation analysis was conducted between these factors and the active components of C.pilosula to identify the key ecological influences that drive the accumulation of active compounds.Results showed that network pharmacology analyses indicated that the active components of C.pilosula,including lobetyolin,syringin,and tangshenoside I,bind to targets and exhibit antioxidant and antipulmonary fibrosis effects.Differences in the contents of active components across three commodity specification grades were not significant.The contents of active components in C.pilosula showed differences with varying origins,with the most variation observed in soluble sugar content,and notable variations are also observed in the levels of lobetyolin,syringin,and tangshenoside I,which could serve as potential biomarkers for different origins.Additionally,ecological factors influenced the accumulation of C.pilosula’s active components.The contents of soluble sugars and tangshenoside I were positively correlated with temperature and precipitation.Our study evaluated the active components of C.pilosula,and findings show that lobetyolin,syringin,and tangshenoside I have potential efficacy in treating pulmonary fibrosis and oxidative stress.The differences in the quality of C.pilosula across varying commodity specification grades are not significant.The different contents of C.pilosula across varying origins are significant,with soluble sugars and glycosides serving as potential markers for distinguishing C.pilosula from different origins.Moreover,ecological factors drove the accumulation of C.pilosula components.Soluble sugars and tangshenoside I content were particularly influenced by temperature and precipitation.Sand content and electrical conductivity significantly correlated with syringin,whereas organic carbon negatively influenced total flavonoids.This research provides a theoretical basis for the selection of the C.pilosula growing area and lays a foundation for the study of the C.pilosula quality standard. 展开更多

Inulin-type fructan CP-A,a predominant polysaccharide in Codonopsis pilosula,demonstrates regulatory effects on immune activity and anti-inflammation.The efficacy of CP-A in treating ulcerative colitis(UC)is,however,not well-established.This study employed an in vitro lipopolysaccharide(LPS)-induced colonic epithelial cell model(NCM460)and an in vivo dextran sulfate sodium(DSS)-induced colitis mouse model to explore CP-A’s protective effects against experimental colitis and its underlying mechanisms.We monitored the clinical symptoms in mice using various parameters:body weight,disease activity index(DAI),colon length,spleen weight,and histopathological scores.Additionally,molecular markers were assessed through enzyme-linked immunosorbent assay(ELISA),quantitative real-time polymerase chain reaction(qRT-PCR),immunofluorescence(IF),immunohistochemistry(IHC),and Western blotting assays.Results showed that CP-A significantly reduced reactive oxygen species(ROS),tumor necrosis factor-alpha(TNF-α),and interleukins(IL-6,IL-1β,IL-18)in LPS-induced cells while increasing IL-4 and IL-10 levels and enhancing the expression of Claudin-1,ZO-1,and occludin proteins in NCM460 cells.Correspondingly,in vivo findings revealed that CPA administration markedly improved DAI,reduced colon shortening,and decreased the production of myeloperoxidase(MPO),malondialdehyde(MDA),ROS,IL-1β,IL-18,and NOD-like receptor protein 3(NLRP3)inflammasome-associated genes/proteins in UC mice.CP-A treatment also elevated glutathione(GSH)and superoxide dismutase(SOD)levels,stimulated autophagy(LC3B,P62,Beclin-1,and ATG5),and reinforced Claudin-1 and ZO-1 expression,thereby aiding in intestinal epithelial barrier repair in colitis mice.Notably,the inhibition of autophagy via chloroquine(CQ)diminished CP-A’s protective impact against colitis in vivo.These findings elucidate that CP-A’s therapeutic effect on experimental colitis possibly involves mitigating intestinal inflammation through autophagymediated NLRP3 inflammasome inactivation.Consequently,inulin-type fructan CP-A emerges as a promising drug candidate for UC treatment. 展开更多

Due to the severe restrictions of natural conditions and ecological environment,high-altitude mountainous areas usually become the"hard bones"in the battle against poverty.Xueshan Township,Luquan Yi and Miao Autonomous County of Yunnan Province,located in the alpine valley of Jinsha River,is a major township with wide and deep poverty,and the incidence of poverty is up to 45.00%.In recent years,Xueshan Township has insisted on the battle against poverty,made effort to develop the Codonopsis pilosula industry,and successfully developed a road to poverty alleviation through C.pilosula industry,and formed a unique industrial poverty alleviation model by the end of 2018,the incidence of poverty dropped to 0.74%.Based on field survey and interview,this paper analyzes and summarizes the specific practices,main results,practical experience and promotion and application measures of the poverty alleviation model of C.pilosula planting industry in Xueshan Township,in the hope of providing certain reference for the targeted poverty alleviation in similar areas in Yunnan Province and other provinces of China. 展开更多

[Results]In 4 groups of rats,there was no significant difference in the time of sexual maturity and ovarian index; compared with the control group,E_2 level of C. pilosula group and P. ginseng group were higher( P < 0. 05);compared with the control group and P. heterophylla group,the ovarian index of P. ginseng group was higher( P < 0. 05); compared with the control group,the body weight of three administration groups increased much more( P < 0. 05). Codonopsis pilosula(CP),a well-known food medicine homology plant,is commonly used in many countries.In our preliminary study,a series of pyrrolidine alkaloids with high MS responses were detected as characteristic absorbed constituents in rat plasma after oral administration of CP extract.However,their structures were unclear due to the presence of various isomers and the lack of reference standards.In the present study,an MS-guided targeted isolation of pyrrolidine alkaloids of CP extract was performed by ultra-performance liquid chromatography coupled with quadrupole time-of-flight tandem mass spectrometry(UPLC/Q-TOF MS).For data analysis under fast data directed acquisition mode(Fast-DDA),an effective approach named characteristic fragmentation-assisted mass spectral networking was successfully applied to discover new pyrrolidine alkaloids with high MS response in CP extract.As a result,seven new pyrrolizidine alkaloids[codonopyrrolidiums C–I(3–9)],together with two known ones(1 and 2),were isolated and identified by NMR spectral analysis.Among them,codonopyrrolidium B(1),codonopyrrolidium D(4)and codonopyrrolidium E(5)were evaluated for lipid-lowering activity,and they could improve high fructose-induced lipid accumulation in HepG2 cells.In addition,the characteristic MS/MS fragmentation patterns of these pyrrolizidine alkaloids were investigated,and 17 pyrrolidine alkaloids were identified.This approach could accelerate novel natural products discovery and characterize a class of natural products with MS/MS fragmentation patterns from similar chemical scaffolds.The research also provides a chemical basis for revealing in vivo effective substances in CP. 展开更多

[Objective] This study aimed to investigate the inspection methods and quality grading standards for Codonopsis pilosula seeds. [Method] A total of 27 shares of C. pilosula seeds were collected and surveyed from six counties in Shanxi, Gansu and Shaanxi provinces. The inspection methods of various indicators, such as purity, 1 000-grain weight, germination rate, vitality and moisture content, were investigated in accordance with Rules for Agricultural Seed Testing (GB/T 3543.1-7, 1995), to develop the appropriate quality inspection method for C. pilosula seeds. Based on the developed inspection method and procedures, the quality of 27 shares of C. pilosula seed samples from different regions was inspected, thus es- tablishing quality grading standards for C. pilosula seeds with K-center clustering method. [Result] C. pilosula seeds are suitable to be cultivated in cool regions. The optimal germination temperature was 25 ~C; the optimal germination bed was gauze bed; the optimal staining conditions were: staining temperature 40 ~, TTC concen- tration 0.5%, staining time 4 h. On the basis of cluster analysis, the quality of C. pilosula seeds could be divided into three grades according to five indicators, includ- ing the purity, 1 000-grain weight, moisture content, vitality, and germination rate. Specifically, vitality and germination rate were important factors of grading standards due to the significant differences, followed by purity, 1 000-grain weight and mois- ture content. [Conclusion] This study provided an important basis for standardized cultivation and industrial development of C. pilosula. 展开更多

Currently,the mortality rate of malignant tumors ranks second globally,surpassed only by cardiovascular and cerebrovascular diseases.The treatment of malignant tumors poses a formidable challenge to both modern medicine and traditional Chinese medicine(TCM).To date,TCM has developed a substantial foundational theoretical understanding and accumulated significant clinical experience in combating tumors.According to TCM foundational theories,"Qi deficiency"is a critical symptom associated with cancer,and"fortifying the body's vitality while expelling pathogens"is the cornerstone of TCM's approach to tumor treatment and bodily balance.Codonopsis pilosula(CP),a Qi-invigorating herb,is known to invigorat the spleen,benefit the lungs,nourish the blood,and promote bodily fluids.It is often employed as a substitute for ginseng in clinical settings.Prolonged clinical observations have identified key active constituents of CP,such as Codonopsis polysaccharides,isoimperatorin,saponins,lobetyolin,sesquiterpene lactones,and muscone.These ingredients exhibit various therapeutic properties,including anti-tumor,immunomodulatory,anti-infective,antioxidant,and hematopoiesis-enhancing effects.Additionally,when CP is combined with other TCM herbs like Astragalus and Atractylodes macrocephala,it bolsters the body's vital energy and rejuvenates both Qi and blood.CP can be used in combination with chemotherapy agents to mitigate the adverse effects of radiotherapy and chemotherapy.Moreover,CP demonstrates potential in preventing precancerous lesions.This review summarizes recent research findings on the anti-tumor properties of CP,elucidates the anti-tumor effects and molecular mechanisms of its active components,provides a basis for promoting the utilization of CP resources and its active constituents,and offers insights for the research and development of new anti-tumor drugs. 展开更多

Protective effect and its mechanism of codonopsis pilosula polysaccharide on memory consolidation disorder induced by cycloheximide in mice

OBJECTIVE To investigate the protective effect of Codonopsis Pilosula Polysaccharide(CPPS)on improving of the memory consolidation disorder induced by Cycloheximide and its possible mechanisms in mice.METHODS The mice was divided into five groups,as normal control group,cycloheximid model group,piracetam positive control group,CPPS 300 mg·kg^(-1) group,and CPPS150 mg·kg^(-1) group.The mice respectively were given saline,piracetam,and CPPS for 15 d.The memory consolidation disorder model in mice was established by ip.Cyclohexylamine,and orally administered CPPS(300 mg·kg^(-1) or 150 mg·kg^(-1))every day.Then experimental groups were subjected Morris Water Maze test.Western blotting analysis were used to analysis the expression of Ca MKⅡ/CREB signaling pathways.RESULTS Morris water maze experiment showed that cyclohexylamine can cause memory consolidation disorder(P<0.01),and giving piracetam and CPPS(300 mg·kg-1)can improve spatial memory impairment in mice(P<0.05,P<0.01).Western blotting experiment results show that compared with normal control group,Ca MKⅡand CREB contents of brain in model group mice had significant decreased(P<0.001);Compared with model group,Ca MKⅡand CREB contents of brain tissue in piracetam and CPPS groups increased significantly(P<0.05,P<0.01,P<0.001).CONCLUSION Cycloheximide can induce the memory consolidation disorder,and its effect in mice related to Ca MK/CREB signaling pathways.CPPS can improved this memory disorder by influence Ca MKⅡ/CREB signaling pathways. 展开更多

[Objectives] This study was conducted to further explore the yield increasing potential of Codonopsis pilosula, and to seek seedlings suitable for efficient production of C. pilosula. [Methods] The effects of C. pilosula seedlings of different sizes on root yield and commercial quality under different cultivation densities were studied. [Results] The fourth-grade seedlings had the least input, the highest yield of 6 948 kg/hm^2, the largest output-to-input ratio at 18.3, the highest root weight proportion of the second-grade product at 58.1%, and the lowest root weight proportion of the first-grade product at 17.6%. Considering the yield, quality and economic benefits comprehensively, the treatment of the fourth-grade seedlings(seedling diameter≤4 mm) is the best choice, and the suitable plant spacing is 8 cm×22 cm. [Conclusions] The study provided a reference basis for the cost saving and efficiency improvement of C. pilosula production. 展开更多

[Objectives]The paper was to establish in vitro propagation of an excellent germplasm of Codonopsis pilosula"Fengdang".[Methods]The effects of different plant growth regulators on callus induction and redifferentiation were tested by using sterile stem explants in vitro that germinated from seeds of superior"Fengdang"plant.[Results]The optimum medium for callus induction was MS+0.5 mg/L NAA+1.0 mg/L 16-BA.The optimal medium for bud differentiation was MS+0.1 mg/L NAA+0.5 mg/L 6-BA,and that for rooting was 1/2 MS+0.2 mg/L NAA+0.2 mg/L IBA.By using the media mentioned above,the rates of callus induction,bud differentiation and rooting reached 91%,100%and 97%,respectively.[Conclusions]The rapid propagation system of regenerated plants in vitro established in this study lays a foundation for the popularization and industrialized seedling of excellent germplasm of"Fengdang". 展开更多

为了探讨硒化党参多糖(sCPP)对海兰褐蛋雏鸡生长性能,免疫器官和血清抗氧化能力的影响,试验采用硝酸-亚硒酸钠(HNO_(3)-Na_(2)SeO_(3))法制备硒化党参多糖,然后进行饲喂试验。选用1日龄海兰褐蛋雏鸡120只,预饲7 d后随机分成6组,每组4个... 为了探讨硒化党参多糖(sCPP)对海兰褐蛋雏鸡生长性能,免疫器官和血清抗氧化能力的影响,试验采用硝酸-亚硒酸钠(HNO_(3)-Na_(2)SeO_(3))法制备硒化党参多糖,然后进行饲喂试验。选用1日龄海兰褐蛋雏鸡120只,预饲7 d后随机分成6组,每组4个重复,每个重复5只进行正式试验。空白对照组饲喂基础饲粮,党参多糖(CPP)组在常规饲粮基础上添加100 mg/kg CPP,sCPP_(1)、sCPP_(2)、sCPP_(3)组分别在常规饲粮基础上添加50、100和150 mg/kg sCPP,Na_(2)SeO_(3)组则在常规饲粮基础上添加0.3 mg/kg Na_(2)SeO_(3)。试验预饲期7 d,正试期28 d,测定不同处理组雏鸡生长性能、免疫器官指数和血清抗氧化能力。结果表明:(1)硒化党参多糖最佳修饰条件为每500 mg多糖加300 mg亚硒酸钠、反应温度90℃、反应时间6 h,含硒量为3.97 mg/g。(2)与空白对照组相比,sCPP_(3)组平均日增重显著提高了13.9%(P<0.05);sCPP_(2)组脾脏指数显著提高了38.8%(P<0.05);sCPP_(1)组和sCPP_(3)组胸腺指数显著提高了39.2%和40.7%(P<0.05)。(3)与CPP组和Na_(2)SeO_(3)组相比,sCPP各组显著提高了蛋雏鸡T-AOC、SOD、CAT、GSH-Px活性(P<0.05)、显著降低了MDA含量(P<0.05)。综上,饲粮中添加100~150 mg/kg的硒化党参多糖可提高雏鸡生长性能,并增强其免疫功能和抗氧化能力。 展开更多

本试验旨在研究党参多糖(CPP)对牦牛犊牛生长性能、养分消化、瘤胃发酵及微生物区系的影响。采用单因子随机试验设计,选取6月龄左右、平均体重为(72.61±1.35)kg、健康状况良好的牦牛公犊27头,随机分为3组:对照组(CN组,仅饲喂基础饲... 本试验旨在研究党参多糖(CPP)对牦牛犊牛生长性能、养分消化、瘤胃发酵及微生物区系的影响。采用单因子随机试验设计,选取6月龄左右、平均体重为(72.61±1.35)kg、健康状况良好的牦牛公犊27头,随机分为3组:对照组(CN组,仅饲喂基础饲粮)、低剂量组(DSL组,在基础饲粮中添加5 g/d的CPP)和高剂量组(DSH组,在基础饲粮中添加10 g/d的CPP),每组9个重复,每个重复1头牛,单栏饲养。预试期10 d,正试期60 d。结果表明:1)DSH组平均日增重(ADG)极显著高于CN组(P<0.01),终末体重(FBW)和平均日采食量(ADFI)显著高于CN组(P<0.05),且ADFI显著高于DSL组(P<0.05)。2)DSH组中性洗涤纤维(NDF)消化率显著高于CN组和DSL组(P<0.05)。3)DSH组总挥发性脂肪酸(TVFA)浓度极显著高于CN组和DSL组(P<0.01),乙酸(AA)浓度显著高于CN组和DSL组(P<0.05)。4)DSH组Shannon指数、ACE指数和Chao1指数极显著高于CN组(P<0.01),DSL组ACE指数和Chao1指数极显著高于CN组(P<0.01),DSH组ACE指数极显著高于DSL组(P<0.01);瘤胃微生物属水平上,DSH组瘤胃球菌属的相对丰度极显著高于CN组(P<0.01),候选糖单胞菌属的相对丰度显著高于CN组(P<0.05);功能预测分析结果显示,脂质代谢及折叠、分类和降解通路在DSH组极显著富集(P<0.01),辅酶和维生素代谢及萜类和聚酮类代谢通路在DSH组显著富集(P<0.05)。5)相关性分析发现,ADG、ADFI和TVFA、AA的浓度与瘤胃球菌属相对丰度、辅酶和维生素代谢通路呈显著正相关(P<0.05);ADFI和TVFA、AA的浓度与折叠、分类和降解通路呈显著正相关(P<0.05);料重比(F/G)与其他氨基酸的代谢通路呈显著正相关(P<0.05)。综上所述,饲粮中添加CPP能够提高牦牛犊牛的生长性能、NDF消化率、TVFA、AA的浓度,改善瘤胃微生物区系结构。 展开更多

Three new sesquiterpene glycosides,named codonopsesquilosides A C(1 3),were isolated from an aqueous extract of the dried roots of Codonopsis pilosula.Their structures including absolute configurations were determined by spectroscopic and chemical methods.These glycosides are categorized as C_(15) carotenoid(1),gymnomitrane(2),and eudesmane(3)types of sesquiterpenoids,respectively.Compound 1 is the first diglycoside of C_(15) carotenoids to be reported.Compound 2 represents the second reported example of gymnomitrane-type sesquiterpenoids from higher plants.The absolute configurations were supported by comparison of the experimental circular dichroism(CD)spectra with the calculated electronic CD(ECD)spectra of 13,their aglycones,and model compounds based on quantummechanical time-dependent density functional theory.The influences of the glycosyls on the calculated ECD spectra of the glycosidic sesquiterpenoids,as well as some nomenclature and descriptive problems with gymnomitrane-type sesquiterpenoids are discussed. 展开更多

为探究复方仙鹤草肠炎胶囊(compound Agrimonia pilosula enteritis capsules,CAPEC)治疗溃疡性结肠炎(ulcerative colitis,UC)湿热证的效应机制,将小鼠随机分为5组:对照组、模型组、阳性药5-氨基水杨酸(5-aminosalicylic acid,5-ASA)组... 为探究复方仙鹤草肠炎胶囊(compound Agrimonia pilosula enteritis capsules,CAPEC)治疗溃疡性结肠炎(ulcerative colitis,UC)湿热证的效应机制,将小鼠随机分为5组:对照组、模型组、阳性药5-氨基水杨酸(5-aminosalicylic acid,5-ASA)组、CAPEC低剂量(CAPEC-L)组、CAPEC高剂量(CAPEC-H)组,采用高糖高脂饮食、白酒饲喂联合葡聚糖硫酸钠建立UC湿热证小鼠模型。通过酶联免疫吸附实验、实时荧光定量PCR、三重四极杆液质联用系统、组织病理学分析等方法,研究了CAPEC对UC湿热证小鼠模型胆汁中胆汁酸(bile acids,BAs)代谢谱及FXR-SREBP-1信号通路的影响。实验结果显示,与模型组相比,CAPEC-L组及CAPEC-H组小鼠疾病活动指数、血清及结肠组织中IL-6、IL-1β、TNF-α等促炎因子水平均显著降低;肠道炎症、肝脏脂质蓄积及舌组织异常改变明显改善。CAPEC-H组胆汁中BAs谱的异常增高显著降低;肝脏Cyp7a1、Cyp7b1、Cyp27a1、Bsep、Fxr、Shp mRNA水平上调;肝脏Srebp-1、Cyp8b1 mRNA水平下调。本研究证实了CAPEC对UC湿热证的改善作用,并提示该作用与其调控胆汁酸代谢、改善肝脏脂质蓄积和肠道炎症有关,为揭示CAPEC清热燥湿、健脾止泻的效应机制提供了线索。 展开更多

为探讨3种不同成分的腐植酸类肥料对党参生长和品质的影响效果,并筛选出效果最佳的产品,以潞党参为研究对象,设置清水(CK)、腐植酸类肥料Ⅰ(T1)、腐植酸类肥料Ⅱ(T2)和腐植酸类肥料Ⅲ(T3)4个处理,研究在花期叶面喷施腐植酸类肥料对党参... 为探讨3种不同成分的腐植酸类肥料对党参生长和品质的影响效果,并筛选出效果最佳的产品,以潞党参为研究对象,设置清水(CK)、腐植酸类肥料Ⅰ(T1)、腐植酸类肥料Ⅱ(T2)和腐植酸类肥料Ⅲ(T3)4个处理,研究在花期叶面喷施腐植酸类肥料对党参产量和品质的调控效应,并对不同腐植酸类肥料处理下的党参叶片光合特性、碳氮代谢酶活性、生长和品质指标进行综合评价。结果表明,3种腐植酸肥料均能促进党参的生长,提高光合效率、代谢酶活性,进而提高产量;同时,对次生代谢物的累积也有不同程度的促进作用。与CK相比,T1、T2和T3处理的产量分别显著提高37.4%、41.1%、54.9%;T3处理的炔苷和紫丁香苷含量分别较CK显著增加了10.4%和6.7%,T2和T3处理的白术内酯Ⅲ含量分别较CK显著提高了11.6%和20.2%。主成分分析结果显示,T3处理对党参生长和品质的提升效果最佳。因此,花期喷施腐植酸类肥料能通过提高根系活力、光合效率和碳氮代谢酶活性来促进党参的生长和次生代谢物的累积,其中腐植酸肥料Ⅲ的促进效果最佳。研究结果为潞党参高产优质栽培提供了一种新途径。 展开更多

本研究以党参为试验材料,通过设置不同用量的化肥与γ-聚谷氨酸(γ-PGA)配施的处理,分析党参生长指标、土壤理化性质和微生物群落的变化,探讨γ-PGA与化肥配施对党参生长的影响及其可能的作用机制。结果显示:与不施用化肥和γ-PGA的对照... 本研究以党参为试验材料,通过设置不同用量的化肥与γ-聚谷氨酸(γ-PGA)配施的处理,分析党参生长指标、土壤理化性质和微生物群落的变化,探讨γ-PGA与化肥配施对党参生长的影响及其可能的作用机制。结果显示:与不施用化肥和γ-PGA的对照(CK)相比,只施用γ-PGA的处理(T1)的党参株高、根直径、根长、根干重均无显著差异,但当γ-PGA与化肥配施后,党参的根干重和根冠比较T1处理显著(p<0.05)增加,增幅分别为4.10%~21.92%、2.45%~17.48%。γ-PGA与适量化肥配施显著提高了土壤的铵态氮、速效钾和有效磷含量,但会降低土壤的pH值。在土壤微生物方面,γ-PGA与适量化肥配施可显著增加土壤中的氨化细菌和好气性自生固氮菌的数量,增强土壤对有机氮的矿化作用和固氮能力。综上,我们认为,γ-PGA与适量化肥配施可通过改善土壤理化性状、促进有益微生物生长、改善根际土壤微生态环境等来促进党参生长。 展开更多

Objective To identify reliable reference genes(RGs) for normalization of real-time PCR(RT-q PCR) data in Codonopsis pilosula. Methods The expression profiles of 10 candidate RGs(GAPDH, ACT, α-TUB, β-TUB, UBQ, CYP, EF-1α, NAC, F-box and PP2A) were examined in C. pilosula during the phenological period. The raw materials examined included roots, stems, leaves, and flower buds at flowering and boll-forming stages, five growth stages of untreated and treated roots with plant growth retardant. The best-suited RGs were accessed using ge Norm, NormF inder, Best Keeper, and Ref Finder algorithms. Results The best-ranked references genes differed across the samples. GAPDH and PP2A were the most suitable for expression analysis in untreated tissues while GAPDH, α-TUB, and PP2A were ranked as the three most stably expressed genes in untreated roots, while NAC and CYP were the most stably expressed genes in stressed(i.e., treated) roots. The expression of UGPase, a key enzyme for CPP biosynthesis, was determined to further validate the selected RGs. Conclusion A total of 10 RGs can be used as reference genes of C. pilosula, however the appropriate one should be used as it may chance. 展开更多
