摘要 目的测定脑血管病人血浆血管内皮生长因子(VEGF)水平及其与病情轻重的关系,探讨VEGF对脑缺血再灌注损伤的作用。方法用ELISA法测定脑血管病人的血浆VEGF水平,采用线栓法做鼠大脑中动脉缺血再灌注模型,比较缺血对照组和VEGF处理组梗死体积和脑组织含水量的差异。结果脑梗死及脑出血患者急性期血浆VEGF水平明显升高(P<0.05),脑梗死患者在患病3周后VEGF较急性期还高(P<0.05),而脑出血患者则较急性期下降(P<0.05),VEGF水平与病情轻重成正比(P<0.05),应用VEGF组脑梗死的体积和脑细胞含水量较对照组明显降低(P<0.05)。结论脑缺血诱导VEGF的产生,而VEGF能减小梗死体积,减轻脑水肿。 Objective: To investigate the plasma level of VEGF in patients with brain vascular disease and different level between slight and severe cases. Then investigate the effect of VEGF on MCAO. Methods: We investigated the plasma level of VEGF of 70 patients with brain vascular diseases by ELISA method. Then we made rat middle cerebral artery occlusion (MCAO) model and divided them into two groups, one was given VEGF through tail vein and the other was ischemic control group. We investigated the infarction volume and brain water content rate between these two groups. Results: The VEGF level increased in patients with cerebral thrombosis and brain hemorrhage (P<0.05). After 3 weeks of cerebral thrombosis, the VEGF increased more that at acute stage (P<0.05) but it decreased in brain hemorrhage (P<0.05). The VEGF level is in direct ratio with patients′ state of illness (P<0.05). VEGF treated rats had smaller infarction volume and less brain water content rate than ischemic control (P<0.05). Conclusions: VEGF is induced by cerebral ischemia and it can decrease the infarction volume and cerebral edema.
