羟乙基淀粉对内毒素感染大鼠中性粒细胞功能的影响及分子机制 被引量:1

摘要 检测羟乙基淀粉 (HES 2 0 0 / 0 .5 )对内毒素腹腔感染大鼠中性粒细胞 (PMN)功能的影响及其分子机理 .雄性Wistar大鼠注射内毒素 (LPS ,6mg/kg ,ip)以造成感染模型 .LPS注射后1min ,4种不同剂量的HES :3.75 ,7.5 ,15 ,和 30mL/kg ,分别由右侧颈静脉持续输注 .LPS注射后 2h及 4h分别检测 :循环血PMN核转录因子kappaB (NF_κB)水平 ,PMN表面CD11b表达 ,和多种组织中PMN浸润情况 .HES (3.75和 7.5mL/kg) ,能降低LPS引起的NF_κB激活和肺、肝、心诸脏器PMN浸润增高 .4种不同浓度的HES剂量依赖性地减少CD11b表达 .HES对感染时PMN的激活和浸润有抑制作用 ,这种作用的产生与它对NF_κB的抑制有关 . The purpose of this study is to examine the effects of hydroxyethyl starch(HES200/0.5)on neutrophils activation and explore the underlying mechanisms.Sepsis was induced in male Wistar rats via injection of lipopolysaccharide(LPS,6mg/kg,ip).HES was infused via the right external jugular vein1minute after LPS challenge at four doses:3.75,7.5,15,and30mL/kg.Nuclear factor kappa B(NF-κB)activation in neutrophils,expression of CD11b on the blood neutrophil cell surface,and neutrophil sequestration in multiple organs were examined2hours or4hours after LPS challenge.Treatment of rats with HES(3.75and7.5mL/kg)prevented LPS-induced NF-κB activation and reduced neutrophil sequestration in lungs,livers,and hearts.Four different doses of HES decreased CD11b expression in a dose-dependent manner.HES could attenuate the neutrophils activation in endotoxic rats.This effect was due to its inhibition of NF-κB activation.
