摘要
研究雷公藤内酯醇对T淋巴细胞中核因子 κB活力及其抑制分子IκB的影响 ,进一步阐明雷公藤内酯醇免疫抑制作用的分子机制 .利用凝胶移率实验 (EMSA)检测不同浓度的雷公藤内酯醇对于普通培养状态下和同时使用PMA/PHA激活的Jurkat细胞中NF κB活力的影响 ,并以逆转录 半定量PCR方法检测了雷公藤内酯醇处理后两组Jurkat细胞中IκBα 的mRNA水平的改变 .研究发现 :①在普通培养状态的Jurkat细胞核内存在一定活力的NF κB ,使用PMA/PHA处理可使NF κB活力显著升高 ,雷公藤内酯醇可以降低两种培养状况下的Jurkat细胞中NF κB活力 ,并以激活状态下更为显著 ;②雷公藤内酯醇可以上调Jurkat细胞中IκBαmRNA的表达水平 .雷公藤内酯醇上调IκBα 基因转录有可能是其抑制细胞内NF
Triptolide is the most potent ingredient for the immunosuppressive effect of Tripterygium Wilfordii Hook f, but its molecular mechanism is still poorly understood. NF κB, a critical regulator for inducible expression of multiple genes involved in inflammatory responses, may be a target for triptolide mediated immunosuppression. Here we measured the activity of NF κB in human T lymphocyte cell line Jurkat cell with EMSA. PMA/PHA treatment was found to induce NF κB activation rapidly. Treatment of triptolide could decrease the activity of NF κB in both PMA/PHA stimulated and non stimulated Jurkat cells. This effect of triptolide was in a dose dependent manner and more obvious in the stimulated cells. IκBα was an inhibitor of NF κB; gene expression was detected by semi quatitative RT PCR in Jurkat cells with different treatment of triptolide. It was found that triptolide could promote the mRNA transcription of IκBα, and it may partly account for the inhibitory effect of triptolide on the activation of NF κB.These results demonstrate that triptolide is a potent inhibitor of NF κB activation, and thus may explain some of the known biological properties of triptolide.
