摘要
目的 研究羟乙基淀粉溶液 (HES 2 0 0 /0 5 )对内毒素腹腔感染大鼠炎症反应的影响 ,并探讨分子机制。方法 雄性Wistar大鼠随机分为对照组、内毒素组 (LPS 6mg/kg)、LPS +HES组 (HES 3 75、7 5、15、30mL/kg)及HES对照组 (30mL/kg)。分别于LPS注入后 4h检测血浆肿瘤坏死因子 (TNF -α)和细胞因子诱导的中性粒细胞趋化因子 (CINC)浓度 ,2h检测循环血单个核细胞核转录因子kappaB(NF -κB)水平。结果 大鼠内毒素感染时血浆TNF -α和CINC浓度及单个核细胞NF -κB活性明显上升 (P <0 0 5 )。 3 75和 7 5mL/kgHES能明显降低TNF -α和NF -κB水平 (P <0 0 5 ) ;3 75、7 5和 15mL/kgHES能明显降低CINC水平 (P <0 0 5 )。结论 较低剂量HES具有抑制内毒素腹腔感染大鼠炎症反应的作用 ,这种作用的产生与其对NF -κB的抑制有关。HES对感染状态有保护作用。
Objective To examine the effects of hydroxyethyl starch (HES 200/0.5)on inflammatory response in endotoxic rats and explore the underlying mechanisms.Methods Male Wistar rats were randomly divided into 7 groups as control,LPS(6 mg/kg),LPS plus HES (3.75,7.5,15,30 mL/kg),or HES (30 mL/kg)alone groups.Plasma tumor necrosis factor (TNF)-α and cytokine-induced neutrophil chemoattractant (CINC) level was measured by ELISA 4 hours after LPS challenge.Nuclear factor kappa B(NF-κB)activation in blood monocytes was measured with electrophoretic mobility shift assay(EMSA) 2 hours after LPS challenge.Results Plasma TNF-α,CINC,as well as NF-κB activity in blood monocytes in LPS challenged rats had increased level as compared with controls(P<0.05).HES,at doses of 3.75 and 7.5 mL/kg,significantly reduced LPS- induced increases of TNF-α level and NF-κB activation(P<0.05);while at doses of 3.75,7.5 and 15 mL/kg,significantly reduced CINC level (P<0.05).Conclusion Lower doses of HES could attenuate the inflammatory response in endotoxic rats.This effect was due to its inhibition of NF-κB activation.HES had beneficial effect on inflammatory situations.
