α_1肾上腺素受体亚型对心肌细胞DNA合成的作用

摘要 用新生乳鼠心肌做原代细胞单层培养后均加入心得安(Propranolo)。分为4组:对照组;去甲肾上腺素(NE)组;NE+CEC组;NE+Nifedipine组。加入~3H-胸腺嘧啶核苷后测放射性计数。结果表明,CEC阻断α_(1b)亚型受体后,不影响NE刺激心肌细胞DNA的合成;Nifedpine阻断α_(1a)亚型受体后,完全阻断了NE的作用。提示,α_(1a)亚型受体可能是交感-儿茶酚胺刺激心肌肥大形成的机理之一。 Neonatal rat cardiac myocytes were monolayerly cultured in the DMEM medium. Forty-eightbours later the agonists and/or antagonists were added in a total volume of 500 ul, in the presence ofpropranolol. The experiments were divided into four groups: the control group; norepinephrine (NE)group; NE+CEC group; NE+Nif. group. After 24 hours the cells were harvested and the radio acti-vity (~3H-TdR) was measured. The results showed that the CPM increased in the NE group comparedwith the control group (p<0.001); the effect of NE was not influenced by CEC, but was totallyblocked by Nif. These suggested that the DNA synthesis in the myocardium cells stimulated by NEwas associated with α_1. adrenoceptor and without α_(1b) adrenoceptor.
