摘要
以猪脑为原料,分段盐析提取后,经过Sephadex-G75凝胶过滤层析,CM-cellulose离子交换层析和反相HPLC,得到反相HPLC单一峰型的神经肽Y。圆二色吸收光谱分析表明:神经肽Y分子中α-螺旋结构的稳定性高度依赖于环境的pH条件。
NPY was isolated from acid extracts of porcine brain. After (NH4)2SO4 fractionation, NPY was purified from the resolubilized 80% -saturation-(NH4 )2SO4 peptide precipitate by Sephadex-G75 gel filtration chromatography, CM-cellulose cation-exchange chromatography and reverse-phase high-pressure liquid chromatography. Circular dichroic spectrum reveals that stabilization of a-helix in NPY is highly dependent on pH of the environmental solution.
