以DNA多聚酶为靶点筛选抗癌药的新方法的初步探讨 被引量:6

摘要 作者借鉴文献方法,从小鼠艾氏腹水癌细胞中提取DNA多聚酶粗酶液,实验观察了十四种已知抗癌药和正在研究中的药物对DNA多聚酶活性的影响,初步建立了以DNA多聚酶为靶点的抗癌药筛选方法。实验表明,能够抑肋DNA多聚酶的药物可能具有抗癌作用。米托蒽醌已在临床肿瘤化疗中广泛使用,对DNA多聚酶有很强的抑制作用,文献尚未见报道。我室研究表明:龙井茶提取物和毛叶茶是取物对小鼠艾氏腹水癌实体型和小鼠网织细胞肉瘤有一定程度的抑制作用。本文证明它们对DNA多聚酶有一定程度的抑制作用。 Referred to method described in literature, DNA polymerase was extracted from Ehrlich ascites carcinoma (EAC) in mice. Effects of fourteen conventional and investigational anticancer drugs (50μg/m.l) on DNA polymerase were tested. We considered that an anticancer drag screening method using DNA polymerase as a target may be established. the obtained results indicated that drugs which could inhibit DNA polymerase may possesses anticancer activities. Mitoxantrone is widely used in clinical canoar chemotherapy, its strong inhibition (100% inhibition at 50μg/ml) on DNA polymerase was observed, which has not been described in the literature . Inhibition of extract of Camellia Ftilophylla Sinensis annd Camellia Ptilophylla Chang on DNA polymerase (39.0%and 87.1% inhibition at 50μg/ml, respectively ) was found. Both drugs were shown (in our laboratory) to inhibit the growth of ESA and L2 tumors.
