摘要
本文用两种悬浮介质,PBS与15%PVP的四种不同类型的血样,用北京医科大学与北京地质仪器厂共同研制的激光衍射仪进行了测量,并作了系统对比,结果表明:(1)对每一种血样,用PBS悬浮介质与PVP悬浮介质所测得的结果定性一致,而PBS缓冲液中所受到的切应力要比PVP中低得多,因而更接近生理条件下的应力。(2)用甲醛固化在PBS切变流场中不同切变率下的红细胞,显微镜下观察证明红细胞在切变率低至γ=20s^(-1)下仍发生变形,这与前人(如R.Hardeman等)所得结论不同。因此,我们认为在我国条件下用激光衍射仪观测红细胞变形性时完全可以用价格低廉的PBS悬浮介质代替价格昂贵的高粘度悬浮介质(如FVP、右旋糖酐等)。
<ABSTRACT>In order to find a new suspending medium for Ektacytometric meas-urement, systematic comparison of measured rheological prope rties oferythrocytes suspended in different buffer has been carried out. Twokinds of buffer were used, e. g. PBS(0. 12M NaCL, 0.02M NaHPO, 0.005M KHPO, 0. 1 % bovin serum albumin, pH = 7.4, 295mOsm/kg, 0.89cp)and PVP (PVP 40000 15%, 0.02M Na2HPo, 0.005M KHPO, pH = 7.4,295mOsm/kg, 61 .71cp ). And four types of erythrocyte were measuredby using both suspending media. that is: normal rabbit's RBC, rabbit'sRBC hardened by formalin with different concentrations, rabbit's RBCtreated by trypsin and human erythrocyte sampled from patients. Undertesting microscope and scanning electron microscope, photographs of er-ythrocytes rigidizied at constant shear rate were observed. Results showthat (1) For each of the four types of erythrocytes, measured deforma-tion index (DI ) --shear rate (r) curves in PBS are similar qualitativelyto those measured in PVP. (2 ) Microscope and scanning electron micros-cope photographs show that erytrocytes indeed deformed when the shearrate of flow field is as low as = 20s-1. It supports the measured resultsby using PBS as suspending medium and the fact shows that, as suspen-ding medium for Ektacytometric measurement, PBS would be better thanPVP, especially in low shear rate area.Therefore, we believe that for Ektacytometric measurement, we sug-gest to use PBS instead of PVP as suspending medium since PBS is muchcheaper than PVP.
