摘要
应用 PVC-ELISA 法,用广州管圆线虫6种抗原对实验感染大白鼠血清中特异性抗体的检测效果进行了初步比较观察。以广州管圆线虫第三期幼虫、颅内期幼虫、雌虫、雄虫与雌雄成虫制备的5种粗抗原及经 Sephadex G200过滤之成虫“纯”抗原的第一和第二蛋白峰,检测感染后第8周鼠血清,其 PVC-ELISA OD 均值分别为0.29、0.29、2.23、1.17、1.64、0.37及0.52,此等 OD 均值与正常鼠血清 OD 均值之比值依次为2.23、2.23、14.87、8.39、11.71、4.63及5.87。结果显示,在所用5种粗抗原中,3种成虫粗抗原作 PVC-ELISA 抗原的敏感性比第三期幼虫和颅内期幼虫粗抗原较高,而雌虫粗抗原的敏感性又较雄虫粗抗原好;成虫‘纯’抗原的敏感性比上述3种成虫粗抗原略低而优于2种幼虫粗抗原。检测感染后第3周大鼠血清,亦显出雌虫粗抗原的血清学活性较颅内期幼虫粗抗原稍高。雌虫和颅内期幼虫粗抗原与感染旋毛虫鼠血清存在明显交叉反应,而成虫‘纯’抗原特异性较高。
The comparison of the sensitivity and specificity of six antigens prepared from A.cantonensis for detection of A.cantonensis antibodies in infected rat sera by PVC-ELISA method has been studied preliminarly.The sera from rats infected with T.spiralis also reacted with 3 kinds of A.cantonensis antigens. The preliminary results showed that among five crude antigens from A.can- tonensis used,three from the adult worms were more sensitive than those prepared from the thirestage larvae and juvenile worms of A.cantonensis recovered from rat brains.The crude antigen from female worms was found to have better sensitivity for antibody detection than those from the male adults.This study also revealed that cross reaction occurred between T.spi- rails and A.cantonensis.Among three A.cantonensis antigens tested,the Sephadex G-200 “purified” antigen from adult worms had better specificity in the PVC- ELISA detection of specific antibody.
