正常不同鼠龄及再生肝的肝细胞DNA含量分析 被引量:3

摘要 用细胞分光光度技术定量测定细胞学涂片中单个肝细胞DNA含量的方法,观察了不同龄正常大鼠及大鼠部分肝切除后再生肝的肝细胞DNA含量情况。结果表明:正常大鼠出生后,随着年龄的增长肝细胞核倍体逐渐从二倍体向四倍体发展;2/3肝切除术后48h,二倍体细胞和双核细胞明显减少,四倍体细胞增多,显示在增生刺激下促进了肝细胞多倍体化过程。最后,对肝细胞多倍体化的形成及其生物学意义进行了讨论。 Suspension of intact liver cells were prepared from rats at different age and liver regeneration. The isolated hepatocytes were smeared. The number of nuclei per cell was determined and the DAN—Content of each single nucleus was measured by microspectrohptometer at a wavelength of 560nm. The transformation of the diploid population of hepatocytes in newborn animals into a polyploid one took place during their pastnatal growth. Analysis of hepatocytes from rats undergoing liver regeneration following two—thirds partial hepatectomy revealed that there were an increase in tetraploid cells and a decrease in diploid and binuclear ones. The polyploidization appeared to be temporarily、 accelerated under the stimulus of proliferation.
