摘要
目的观察不同状态的kupffer细胞(KC)对肝细胞再生的影响,探讨肝脏再生启动的机制。方法复制肝硬变大鼠肝部分切除术模型,体外培养肝细胞和KC,测定其上清液(TNF-α)和表皮生长因子(EGF)等细胞因子含量和观察肝细胞DNA、蛋白质的合成。结果肝细胞加上C-PH组的KC上清液时,肝细胞DNA和蛋白质合成能力明显降低,而肝细胞加上正常对照组KC的上清液时,肝细胞DNA和蛋白质的合成功能均明显增加。C-PH组肝细胞在术后早期EGF分泌量低.而TNF-α含量高,肝细胞再生启动延迟。结论 KC通过分泌细胞因子刺激和抑制肝细胞生长,但受损的KC使肝细胞再生启动延迟。
Objective To observe effects of Kupffer cells in diffrent conditions on liver regeneration,and to fur- ther probe mechanism of hepatocyte regeneration "start",Methods The cirrhotic rats with partial hepa- tectomy were used as an animal model. Hepatocyts and Kupffer cells were cultured in vitro,and TNF-α, EGF and other cytokines in the cultured cell supernatant were determined. The DNA and protein synthe- sis of the rat hepatocytes were observed. Results When the hepatocytes were cocultured with the Kupf- fer cell supernatant of the cirrhotic rats after operation,the hepatocyte DNA and protein synthesis func- tions was obviously decreased. However,the liver ceils were cocultured with Kupffer cell supernatant of the normal rats,these functions significantly increased. In the early phase after operation,the EGF level of the hepatocyte was lower,but its TNF-α was higher. Conclusion Kupffer cells can inhibit and stimulate liver regenertion with secreting cytokines,but injured Kupffer cells can delay liver regeneration "start".
