摘要
应用40个10碱基随机引物对长江干流金口和瑞昌江段的青鱼(Mylopharyngodonpiceus)群体以及长江水系的湘江青鱼群体进行随机扩增多态DNA(RAPD)分析,其中29个引物能扩增出1~10条清晰的RAPD带,有14个引物(占全部引物的35%)在青鱼不同个体的扩增产物中有多态性DNA片断。结果表明,青鱼金口、瑞昌、湘江3个群体的遗传相似度的平均值分别为0.9524、0.9612、0.9578,3个群体的Shannon遗传多样性值分别为0 2057、0.1483、0.1821。群体间的遗传相似度依次为金口-湘江:0.9487,瑞昌-湘江:0 9503,金口-瑞昌:0.9512,群体间的遗传相似度低于群体内的遗传相似度,遗传变异度则相反。3个群体的遗传分化指数为14.6%。
Genetic analysis of three populations of black carp ( Mylopharyngodon piceus) from Jinkou,Ruichang and Xiangjiang within the middle reaches of the Yangtze River was conducted by using randomly amplified polymorphic DNA assay.Fourty 10-mer primers were used, of which, 11 produced nothing, and 29 did 1~10 DNA bands, respectively. Fourteen primers produced polymorphic DNA bands. Genetic analysis showed that the genetic similarity and the Shannon's index within Jinkou, Ruichang and Xiangjiang populations were 0.952 4,0.961 2 ,0.957 8 and 0.205 7,0.148 3,0182 1 separatively. The genetic similarity between populations of Jinkou-Xiangjiang, Ruichang-Xiangjiang and Jinkou-Ruichang were 0.948 7, 0.950 3 and 0.951 2 respectively. The genetic similarities in the three populations were higher than that between them. The coefficient of gene differentiation (Gst) was 14.6%.
