通心络抑制缺氧诱导的血管内皮细胞凋亡及机制研究 被引量:24

The Efficacy and the Mechanism of Tong Xin Luo on the Inhibition of Endothelial Cell Apoptosis Induced by Hypoxia

摘要 目的 :研究通心络对缺氧诱导的血管内皮细胞凋亡及Caspase 3活性的影响。方法 :培养的牛主动脉内皮细胞分为三组 :正常对照组 ,缺氧组和缺氧加通心络组 ,2 4h后检测凋亡细胞数并计算凋亡百分比 ,以及Caspase 3蛋白的表达与活性。结果 :三组细胞凋亡百分比分别为 7± 0 12 % ,2 8 85± 2 4 6 %和 11 4 8± 1 4 % (P <0 0 5 ) ;通心络对缺氧后血管内皮细胞的Caspase 3蛋白表达无影响 ,但可抑制其活性增高 (41 2± 5 5U/mlvs 6 7 8± 7 2U/ml,P <0 0 5 )。结论 :通心络减少缺氧所致血管内皮细胞凋亡与抑制Caspase 3的活性有关。 Objective:Determine the efficacy of Tong Xin Luo(TXL)on the apoptosis and the activity of Caspase-3 in the endothelial cell under hypoxia.Method:the Bovine aortic endothelial cells(BAEC) were divided into three groups and incubated with normal medium,hypoxia medium,hypoxia medium+TXL(50ug/ml)for 24h respectively.Apoptosis were measured with TUNEL.Western blot and the kit of Caspase-3 activity determination were used to measure the expression and the activity of Caspase-3 in BAEC.Result:TXL can decrease the percent of apoptotic cells in BAEC induced by hypoxia with mark significance (11.84±1.4% vs 28.85±2.46%, P <0.05).Although TXL had no effect on the expression of Caspase-3 after hypoxia stimulation,TXL could abolished the increase of the activity of Caspase-3 in BAEC induced by hypoxia(41.2±5.5 VS 67.8±7.2 U/ml, P <0.05).Conclusion:The inhibition of the activity of Caspase-3 might be the main mechanism of TXL decrease endothelial cell apoptosis induced by hypoxia.
