摘要
柱切换 HPL C法同时测定人血浆中阿莫西林和克拉维酸浓度。血浆样品加头孢羟氨苄为内标 ,用乙腈沉淀蛋白及二氯甲烷反洗 ,水相进样后经在线净化 ,由流动相洗脱切入 C1 8分析柱进行分离测定。阿莫西林和克拉维酸线性范围分别为 0 .5~ 16和 0 .2 5~ 5 μg/ ml,净化回收率分别为 92 .8%~ 95 .2 %和 77.3%~80 .8% ,方法回收率分别为 94 .0 %~ 96 .1%和 88.9%~ 97.6 % ,日内 RSD分别为 0 .6 3%~ 1.6 1%和 1.4 5 %~ 2 .94 % ,日间 RSD2 .30 %~ 5 .31%和 5 .0 2 %~ 7.37%。本法准确可靠 。
A column switching HPLC method has been developed for the simultaneous determination of amoxicillin and clavulanate in human plasma. The plasma proteins were precipitated by acetonitrile and the supernatant was backwashed with methylene chloride. Cefadroxil was added before as an internal standard. After the aqueous layer injected was on line washed, the residues were eluted by the mobile phase onto analytical column to separate and determine. For amoxicillin and clavulanate, the calibration linear range were 0 5~16 and 0 25~5μg/ml; the recoveries of cleanup were 92 8%~95 2% and 77 3%~80 8%; the recoveries of method ranged from 94 0%~96 1% and 88 9%~97 6%; intra day RSD were 0 63%~1 61% and 1 45%~2 94%; inter day RSD were 2 30%~5 31% and 5 02%~7 37% respectively. This method is accurate and reliable. It has been used to the determination of amoxicillin and clavulanate in human plasma for clinical pharmacokinetic studies.
