人血清对大蒜新素细胞毒性及其抗人巨细胞病毒效应影响的实验研究 被引量:2

摘要 目的 探讨人血清干预对大蒜新素细胞毒性及其抗人巨细胞病毒 (HCMV)效应的影响。方法 将系列浓度大蒜新素与不同浓度血清共同作用于人胚肺成纤维细胞 ,观察细胞增殖活力 (MTT法 )、细胞形态和超微结构及HCMV蚀斑抑制率的改变。结果 ①不加血清时 ,细胞对大蒜新素最大耐受浓度 (MTC)为 9 6 μg/ml;加入 2 0 %灭活血清后 ,MTC增至 2 1μg/ml。②血清干预前 ,2 1μg/ml大蒜新素使 6 0 %细胞坏死 ,胞质与胞核疏松混浊、呈筛状 ,内质网、线粒体弥漫性肿胀空泡变明显 ,异染色质减少 ;血清干预后 ,细胞形态与超微结构均明显改善 ,尤其在干预后 4 8h ,细胞超微结构与正常细胞相似。③ 2 0 %血清干预提高细胞对大蒜新素的最大耐受浓度 ,后者对HCMV抑制率由干预前的 6 5 92 %提高至 82 98%。结论  2 0 %血清干预可明显改善大蒜新素的细胞毒性、保护细胞超微结构 ;增加大蒜新素用量 ,进而提高其抗HCMV效应。 Objective To investigate the influence of human serum on the cytotoxicity and anti HCMV activity of allitridin. Methods Serial concentrations of allitridin, different concentrations of human serum and human embryo lung fibroblasts (HEL) were cultured together. The proliferative activity of HEL (MTT colorimetric assay), the changes of cell morphology and ultrastructures and inhibitory efficacy of allitridin against HCMV were observed.Results The maximum tolerance concentration (MTC) of HEL to allitridin treatment was 9 6 μg/ml before the serum intervention and increased up to 21 μg/ml after adding 20 % deactivated serum into the medium. With only 21 μg/ml allitridin treatment, 60 % of HEL cells developed cellular necrosis, in which cytoplasm and nucleus were rarefied and turbid and looked like sieve pore, endoplasmic reticulum and Golgi body showed suffusive swelling and vacuolation and heterochromatin decreased. After the 20 % serum intervention, the cell morphology maintained normal under the light microscope and the ultrastructures had a little changes and then became similar to normal cells under the electron microscope at 48 h after the intervention. Moreover, the inhibitory efficacy of allitridin against HCMV was increased from 65 92 % to 82 98 % because the MTC of allitridin rose after the serum intervention. Conclusion Adding 20 % serum could ameliorate the cytotoxicity of allitridin and protect ultrastructures of HEL. The inhibitory efficacy was improved by increasing the concentration of allitridin.
