摘要
目的 了解新发现的脑红蛋白基因在人体不同细胞组织中的分布表达。方法 人 13种组织总RNA ,各等量取总RNA 2 μg ,逆转录成cDNA。取上述逆转录产物cDNA 1μl作为模板做PCR。等量取上述每个标本总RNA 2 μg ,逆转录成cDNA。取PCR产物行琼脂糖凝胶电泳ImageJ软件定量分析电泳结果。 结果 1.NGB、GAPDH扩增后的产物片段与设计相符。 2 .人 13种组织中都有NGBmRNA表达 ,各组织中NGBmR NA扫描的相对光密度 (A)值胎肾最高 ;其次为肝组织、大脑皮层、全脑、胎肺、回肠慢性炎症、小脑、胎胸腺、膀胱癌、乳腺癌、Hela S3细胞、肝癌、胎心 ,提示NGBmRNA在脑组织中表达量相对较高 ,胎肾和正常人肝组织的表达亦很高。结论 人体不同细胞组织中都有NGBmRNA表达 ,但不同的组织其表达水平不同 ,提示NGB基因并不是神经系统所特有的 。
Objective To study the expressions of neuroglobin gene in cellular tissues of human body.Methods Total 13 species tissue RNA, superscription RNA 2 μg of every tissue was reversed into transcription cDNA and 1 μl of cDNA was made into PCR template. Total RNA 2 μg from every specimen equivalently were reversed into transcription cDNA. PCR products experienced agarose gel electrophoresis and electrophoresis and extraviolet-gelatum Image J was quantitatively analyzed. All data were indicated with ±s and treated with Oneway mono agent analysis of variance of stata statistical package,P<0.05 (significant difference).Results 1.NGB and GAPDH product fragments after amplification were in accord with the design.2.There were expressions of NGB mRNA in human 13 species besides brain tissue: foetus kidney,liver tissue,cerebral cortex,brain,foetus lung,ileochronicity inflammation,cerebellum,foetus thymus,carcinoma of bladder,mammary adenocarcinoma,Hela-S3 cellular,hepatocarcinoma,embryocardia.The expressions were higher in foetus kidney and normal person liver tissue.Conclusions There are expression of NGB mRNA in human 13 species cellular tissues,and expressions ere at different levels in those tissues. It is suggested that NGB gene dos not belong to the nervous system only,and NGB mRNA may play a more comprehensive role in human body.
