摘要 目的 探讨CpG在幽门螺杆菌疫苗研究中的佐剂效应。方法 将 60只BALB c小鼠分为 4组 ,分别以H .pylori尿素酶B亚单位 (rUreB) +CPG、rUreB +CT、单独UreB以及PBS进行口服免疫 ,ELISA检测胃、肠、气管冲洗液sIgA以及血清IgG亚类 (IgG1,IgG2a) ,RT PCR差异显示T淋巴细胞IFN γ、IL 4mRNA。结果 ①CpG +rUreB组在胃、肠、气管和血清产特异性抗体水平显著高于PBS组和单独rUreB组 (P <0 0 1) ,与CT +UreBs组差异不显著 (P >0 0 5 ) ;②抗原刺激后CpG +rUreB组T淋巴细胞表达的IFN γ、IL 4mRNA显著高于PBS组和rUreB组。结论 ①CpG可能是H .pylori疫苗研究中有效的粘膜佐剂 ;②CpG +rUreB组诱导的可能是Th1 Th2型免疫应答。 Objective To study the adjuvant effects of CpG motif containing oligodeoxynucleotide (CpG) on Helicobacter pylori ( H. pylori ) vaccine. Methods A total of 60 adult mice were divided randomly into four groups. Mice in groups Ⅰ, Ⅱ, Ⅲ, and Ⅳ were orally immunized with recombinant H. pylori urease B subunit (rUreB) plus CpG, rUreB plus cholera toxin (CT), single rUreB, and phosphate buffered saline (PBS), respectively. The specific secreting IgA (sIgA) and specific IgG subclasses (IgG1 and IgG2a) in gastric mucosa, intestinal mucosa of every mouse, tracheal mucosa were determined by ELISA. The expression levels of T cell cytokine IFN γ and IL 4 mRNA in spleen were detected by RT PCR. Results ① The levels of specific antibody in the gastric mucosa, intestinal mucosa, tracheal mucosa, and serum in group Ⅰ were significantly higher than those in group Ⅲ and group Ⅳ ( P <0 01). ② The expression levels of IFN γ and IL 4 mRNA of T cell in group Ⅰ after stimulation with antigens were significantly higher than those in group Ⅳ and group Ⅲ ( P <0.01). Conclusion CpG may be the optimal mucosal adjuvant of H. pylori vaccine. Th1/Th2 type immune response might be induced by oral immunization with CpG+rUreB.
