摘要
目的 :建立测定人血浆中氯氮平浓度的HPLC法。方法 :血浆经碱化后 ,用OASIS固相萃取小柱提取血浆中氯氮平 ,采用反相高效液相色谱法二极管阵列检测器检测。色谱柱为DikmaDiamonsilC18柱 (2 0 0mm× 4 6mm ,5 μm) ,保护柱为WatersNova PackC18柱 ,流动相为 0 5 %三乙胺溶液 (磷酸调pH =3 0 ) 乙腈 (72∶2 8,V/V) ,流速为 1mL·min-1,检测波长 (λ) 2 15nm。结果 :本方法在 0 10~ 2 0 0mg·L-1范围内线性良好 (r=0 9988,n =6 )。高、中、低浓度加样回收率均在 92 1%~ 10 5 8%之间 ,日内、日间RSD在 4 1%~ 8 6 %之间 ,最低检测浓度为 5 μg·L-1。结论
AIM:To establish a simple,sensitive and rapid HPLC method for determination of clozapine in human plasma. METHODS:The clozapine in plasma samples(1 mL)was processed by Waters OASIS solid-phase extractor. The separation used a reversed-phase Dikma Diamonsil C 18 column(200 mm×4.6 mm,5 μm)with mobile phase consisting of 0.5% triethylamine solution (adjust pH to 3.0 with H 3PO 4)-acetonitrile (72∶28,V/V),the flow rate was 1 mL·min -1,the detective wavelenth was 215 nm. RESULTS:The range of calibration curve was 0.10-2.00 mg·L -1(r=0.998 8, n=6),the limit of detection for clozapine in plasma was 5 μg·L -1. The recovery of low,medium and high concentration were within 92.1%-105.8%. The relative standard deviations of intra-day and inter-day assay were within 4.1%-8.6%.CONCLUSION:The method is rapid,sensitive and efficient which can simultaneously determine the concentration of clozapine in plasma.
