摘要 目的:建立人血清中氯氮平的高效液相色谱法。方法:取血清经乙酸乙酯/二氯甲烷(80:20)萃取,再经浓缩离心后,用反相液相色谱法检测。流动相为甲醇-乙酸铵(0.02 mol/L)(70:30,v/v);流速为1.0 ml/min;检测波长254 nm。结果:在0.05-2.0 mg/L范围内方法线性关系良好(r=0.9995),加标回收率在85.0%-91.6%之间,相对标准偏差为3.80%-4.52%,最低检出浓度为0.01 mg/L(S/N=10);中毒病人发病后3、7、10 h所取血样中,氯氮平浓度分别为1.22、0.53、0.14 mg/L。结论:本法灵敏、快速、简便、准确,适用于人血清中氯氮平浓度监测。 Objective:To establish a method for the determination of clozapine in serum by HPLC and detect clozapine in patient′s serum.Methods:The sample was extracted by acetic ether/dichloromethane(80:20).After being concentrated and centrifuged,the sample was analyzed by HPLC.Results:The calibration curve showed good linearity in the range of 0.05~2.00 mg/L.The average recovery was 85.0%~91.6% and relative standard deviations 3.80%~4.52%,the lowest detectable concentration 0.01 mg/L.The concentration of clozapine in serum were 1.22,0.53 and 0.14 mg/L,respectively at 3,7,10 h after onset of poisoned patient.Conclusion:The method is sensitive,accurate,simple and fast.It can be used for determination of clozapine level in serum.
