摘要
以黄色短杆菌TQ9806(Met-+IleL+2-TAr+α-ABr+β-HLr+SGr+Rifr300μg/mL)为出发菌株,经硫酸二乙酯、紫外线分别与氯化锂复合诱变,选育出一株利福平抗性为800μg/mL的L 亮氨酸产生菌TK0303.通过对其摇瓶发酵条件的研究和优化,产量由出发菌株TQ9806的22.7g/L提高至目的突变株TK0303的28.3g/L.
Original strain, Brevibacterium flavum TQ9806 (Met^-+IIe^L+2-TA^r+α-AB^r+β-HL^r+SG^r+Rif^r 300 μg/mL) , was mutated by DES and LiCl, UV and LiCl successively. Then an L-leucine producing strain TK0303 which was resistant to rifampin 800 μg/mL was obtained.Finally the conditions of shake-flask fermentation were studied and optimized. Under these conditions, the mutant could produce higher L-leucine production of 28.3 g/L than 22.7 g/L of TQ9806.
