摘要 目的 :探讨凋亡抑制基因Survivin在人肝癌 (HCC)中的表达及其临床意义。 方法 :应用SABC免疫组化染色技术和逆转录聚合酶链反应 (RT PCR) ,检测 95例HCC及其癌旁组织、8例非肝癌肝组织Survivin蛋白以及 8例HCC组织SurvivinmRNA的表达 ,分析其与病理分级、临床分期和肿瘤大小等临床指标之间的关系。 结果 :HCC组织中Survivin基因蛋白表达率显著高于癌旁组织和非肝癌肝组织 (5 9.77%vs 2 6 .0 0 %和 5 9.77%vs0 .0 0 % ,P <0 .0 1) ;HCC组织中SurvivinmRNA表达阳性率显著高于癌旁组织 (37.5 %vs 0 .0 0 % ,P <0 .0 1) ;Sur vivin蛋白表达与肝癌组织学分级、临床分期、肿瘤大小无相关关系。 结论 :Survivin基因在HCC组织表达上调 ,提示该基因参与了HCC的发生过程 ;Survivin基因表达与组织分化程度无关 ,提示其可能成为HCC特异性诊断指标或基因治疗的新靶点。 Objective:To study the clinical significance of the expression of survivin gene protein in human hepatocellular carcinoma(HCC). Methods: We detected the expression of survivin gene protein in 95 cases of HCC and its adjacent noncancerous tissues and 8 cases of liver tissues by using SABC immunohistochemical staining.The survivin mRNA expression was also detected by using RT-PCR in 8 cases of HCC.The correlation of the expression of survivin gene protein with histologic differentiation, clinical stages and tumor size was studied. Results:The positive expression rate of survivin gene protein in HCC(59.77%) was much higher than the adjacent noncancerous tissues(26.00%) and liver tissues (0.00%) (P<0.01); The positive expression rate of survivin mRNA in HCC(37.5%) was much higher than the adjacent noncancerous tissue(0.00%)(P<0.01);The survivin gene expression did not demonstrate correlation with histologic differentiation, clinical stages, and tumor size by statistical analysis. Conclusion:The up-regulation expression of survivin gene in HCC suggested that survivin participate in carcinogenesis.The fact that there was no significant correlation between survivin gene expression and histologic differentiation suggested that survivin is likely to be identified as a special index in diagnosis and a potential therapeutic target in HCC.
