摘要
目的:研究小鼠肝癌组织中的SP细胞,从而推测肝癌干细胞(CSC)存在的可能性。方法:首先用致癌剂二乙基亚硝胺(DEN)诱导B6C3F1雄性小鼠发生肝癌,在无菌条件下,将肝癌组织消化成单细胞悬液,然后用荧光染料Hoechst 33342对细胞悬液进行染色分析。结果:在原代分离的小鼠肝癌细胞群中观察到SP细胞的存在。结论:实验结果推测,在药物诱导转化的小鼠肝癌组织中很可能存在CSC。
Objective :The tumor, and to speculate the step is to induce liver tumor aims of this experiment is to find side population (SP) cells in mouse liver existence of liver tumor stem cell. Methods:In this experiment, the first in male B6C3F1 mice by carcinogen diethylnitrosamine and obtain the singlely suspened cells at sterile conditions. Then the SP phenotype of these singlely suspened cells were diplayed by dyeing with fluorescent Hoechst 33342. Results:The SP cells were observed in the singlely suspended primary cells of mouse liver tumors. Conclution : The results of this experiment indicated the existence of liver cancer stem cells.
