摘要
建立了应用反相高效液相色谱法测定发酵液中氨苷霉素含量的方法。较佳的分析条件为:色谱柱Hypersil BDS ODS(250×4.6mm I.D.,5μm);流动相为含5.8 mmol/L三氯乙酸和8%(v/v)甲醇的水溶液;检测波长279nm,流速1.0 ml/min。对标准曲线、精密度、准确度及发酵液样品测定的重现性进行了详细的研究。该方法的线性相关系数0.9998,日内变异系数0.05%,日间变异系数0.9%,回收率96.30%。本方法具有准确性高、重现性好、灵敏度高及快速测定的优点,非常适合于发酵液中氨苷霉素含量的测定。
A reverse phase high performance liquid chromatography method for the determination of anganmycin in fermentation broth was developed. Analytical conditions involved use of a Hypersil BDS ODS column (250×4.6 mm i.d., 5μm particle size), with a mobile phase of 5.8 mmol/L trichloroacetic acid and 8% (v/v) methanol and a flow rate of 1.0 ml/min. Wavelength of the UV detector was 279 nm.Standard curves, precision, recovery, and repeatability of broth samples were studied in detail.The linear correlation coefficient for the method was 0.9998, and recovery rate was 96.3% .Coefficient of variation within and between days was 0.05% and 0.9%, respectively.Using this method, anganmycin in fermentation broth can be determined with the advantages of high accuracy, sensitivity, and repeatability.
