亚低温条件下局部脑神经元c-fos蛋白表达变化 被引量:10

Changes of c-fos protein expression in local brain nerve cells after mild hypothermia treatment

摘要 目的:研究颅脑枪弹伤常温下及全身亚低温治疗后脑神经元c-fos蛋白表达的变化。方法:18只杂种犬,随机分为常温组(正常犬温为38.5~39.5℃),亚低温组(31.5~32.5℃)。以德国小口径步枪子弹致伤犬颅脑贯通伤(penetratingcraniocerebralinjury,PCI)模型为对象,采用免疫组化法检测两组脑组织伤后30min,2,3h弹道挫伤区,震荡区及脑干神经元中fos蛋白的表达。结果:全身亚低温治疗3h组弹道挫伤区、震荡区及脑干神经元中fos蛋白阳性数分别为(15.5±1.6),(21.6±2.1),(6.7±1.8)个,较常温对应组弹道挫伤区,震荡区及脑干神经元中fos蛋白阳性数分别为(21.6±5.4),(32.2±1.7),(11.2±1.8)个显著减少(P<0.01)。结论:颅脑枪弹伤后全身亚低温治疗能够抑制脑神经元c-fos蛋白的表达。 AIM:To investigate the changes in the expression of c fos protein in cerebral cortex of canines with craniocerebral gunshot wounds after treatment with normothermia and with mild hypothermia respectively. METHODS:Eighteen canines were randomly divided into normothermic group and mild hypothermic group.Body temperature of the normothermic group was maintained at 38.5-39.5 ℃.In the hypothermia group,the canines were exposed to hypothermia of 31.5-32.5 ℃.The canine model of penetrating craniocerebral injury(PCI)was made by the bullets of the small bore rifle made in Germany.The fos positive neuron expression in the neurons of the two groups was observed with immunohistochemical method in the contusion area and concussion area of the trajectory and the brain stem at 30 minutes,2 and 3 hours after trauma. RESULTS:Three hours later, the numbers of positive expressed c fos proteins in the mild hypothermic group was 15.5±1.6 in the contusion area, 21.6±2.1 in the concussion area and 6.7±1.8 in the brain stem, which was significantly lowered as compared with that in the normothermic group(21.6±5.4,32.2±1.7 and 11.2±1.8,P< 0.01). CONCLUSION:Mild hypothermia after craniocerebral gunshot wounds can restrain the expression of c fos protein in the neurons of brain.
