摘要
目的 :揭示羊栖菜多糖 (SFPS)的抗肿瘤作用机制。方法 :采用流式细胞仪观察羊栖菜多糖对肿瘤细胞周期及细胞凋亡的影响。采用Fluo 3 /AM探针标记 ,激光共聚焦技术观测细胞内 [Ca2 + ]i。结果 :羊栖菜多糖可阻滞SGC 790 1人胃癌细胞由G0 /G1期进入S期 ,升高细胞凋亡指数 (APO % )。可使SGC 790 1细胞内 [Ca2 + ]i先升高然后下降 ,给CaCl2 后 ,[Ca2 + ]i又升高。结论 :羊栖菜多糖通过升高肿瘤细胞内 [Ca2 + ]i启动肿瘤细胞凋亡机制而达到抗肿瘤的作用 ,[Ca2 + ]i升高时Ca2 + 来源于细胞内钙库释放。
Objective: To study mechanism of antitumor a ctivity of Sargassum Fusiforme Polysaccharide(SFPS). Method: The effect on cell cycle and apoptosis was studied with flow cytometry(FCM). Intracellular calcium concentration [Ca 2+ ]i was marked with Fluo-3/AM and measured with laser scanner confocal microscope(LSCM). Result: SFPS inhibited G 0/G 1 stage SGC-7901 from entering to S stage an d increased APO%. The [Ca 2+ ]i showed a transient rise and return to th e original level. The concentration could be raised again by administering CaCl 2. Conclusion:The antitumor effect of SFPS seems to be accomplished through the apoptosis associated with the increase in intracellular calcium concentration. Intracellular stores release the calcium during its acti on.
