摘要
实验发现 ,在 p H8.8的 H3BO3- Na OH介质中 ,镍 ( ) -血清蛋白配合物在单扫示波极谱上 ,在- 0 .6 0 V处 (vs.SCE)会产生吸附性质的催化前波 ,该波二阶导数波高与蛋白质浓度在一定范围内呈线性关系 ,对于牛血清蛋白 (BSA) ,线性范围为 1~ 2 9μg/ m L,检出限为 0 .5 μg/ m L;对于人血清蛋白 (HSA) ,线性范围为 0 .8~2 6 μg/ m L,检出限为 0 .4 μg/ m L。应用该法测定了人血清样品总蛋白含量。研究了实验条件的选择 。
In H 3BO 3-NaOH medium (pH 8.8),the complex of nickel(Ⅱ)-serum albumin yields a polarographic catalytic prewave at about -0.60V (vs.SCE). There is a linear relationship between the second order derivative peak height and the concentration of protein in a certain range. For BSA (bovine serum albumin),the linear range is 1~29μg/mL with the detection limit of 0.5μg/mL, and for HSA (human serum albumin),the linear range is 0.8~26μg/mL with the detection limit of 0.4μg/mL. The method has been used to the determination of total protein content in samples of human serum with satisfaction. The experimental conditions and the mechanism of the method were investigated.
