摘要
为了从红花(Carthamus tinctorius L.)培养细胞中筛选到具有高产α-生育酚性能的细胞变异体,利用细胞平板培养技术,利用HPLC法测定培养细胞中α-生育酚的含量,对200多个由单细胞或小细胞团发育而来的细胞克隆进行了筛选。结果发现,在细胞生长速率和α-生育酚含量上,这些克隆之间存在着显著的差异。在克隆的第一代分析过程中,发现α-生育酚含量由0—138.9μg/gDW变化;细胞生产速率由0.20—0.53gDW/1.d变化。细胞生长速率与α-生育酚含量之间无明显相关性。对一些α-生育酚含量较高的克隆进行了稳定性观察,经近20代继代培养观察后,从中筛选到一个合成α-生育酚较稳定的高产克隆系(CT-289),其α-生育酚平均含量为114.4μg/gDW,是原始株系(15.83μg/gDW)的7.2倍;其生长速率为0.424gDW/1.d,与原始株系(0.417gDW/l.d)相差不大。
Plating culture technique was used for screening high-producing of α-tocopherol variants from the culture cells of Carthamus tinctorius L. The content of α-tocopherol in culture cells was measured by HPLC method. The cell variants were selected from more than 200 clones induced from single cell or small cell aggregates (2-8 cells), and found that these clones were significant difference in a-tocopherol synthesis and cell growth, α-tocopherol contents were varied from 0 to 138.9μg/ gDW and cell growth rates were varied from 0.20 to 0.53 gDW/ l.d at the first generation of clones. There was not a significant correlation between α-tocopherol productivity and cell growth in these clones. The stability of some clones which high-producing α-tocopherol was tested. A more stable high-producing α-tocopherol clone b'ne (CT-289) had been selected through out near 20 passages successive subcultunng. Its mean content of a-tocopherol was 114.4μg / gDW, and was 7.2-fold to theoriginal strain (15.83μg/ gDW). The growth rate of CT-289 was 0.424 gDW/ l.d, and was similar to the original strain (0.417g DW / l.d).
