阿奇霉素对弓形虫病大鼠睾丸组织细胞凋亡影响的实验研究 被引量:6

摘要 目的 检测阿奇霉素对弓形虫病大鼠睾丸组织细胞凋亡的影响。方法 腹腔注射 2× 10 5/ml活的纯化弓形虫速殖子悬液 (CF- 11纤维素纯化 ) 2 m l于 SD大鼠 ,建立大鼠弓形虫病模型 ,用阿奇霉素 2 0 0 mg/ (kg.d)连喂 7d,治疗实验大鼠。 9周后解剖大鼠 ,取睾丸制成睾丸组织单细胞悬液 ,应用流式细胞仪检测其细胞凋亡。同时设有感染组和正常大鼠对照组。结果 感染组和治疗组大鼠睾丸组织细胞凋亡率显著高于正常对照组大鼠睾丸组织细胞凋亡率 (P<0 .0 5 )。治疗组大鼠睾丸组织细胞凋亡率明显低于感染组 (P<0 .0 5 )。结论 弓形虫感染可引起大鼠睾丸组织细胞发生凋亡。但阿奇霉素则可有效地抑制弓形虫感染大鼠睾丸组织细胞发生凋亡 ,可有效地治疗弓形虫感染对大鼠的雄性生殖损害。 Objective To detect the effects of azithromycin on apoptosis of testis spermatogenic cells in rats infected with Toxoplasma gondii. Methods SD rat model of toxoplasmosis was established by ip. injection with 2×10~5/ml KS liver Toxoplasma tachyzoits 2 ml. Rats in the experimental group were treated with 200 mg/kg azithromycin and sacrificed to obtain testis spermatogenic cells. The spermatogenic cells were separated from the testis .The testis spermatogenic cells' apoptosis was detected by flow cytometry(FCM). A group of infected rats and a group of normal rats served as controls. Results Apoptotic rate of testis spermatogenic cells in the infected or treated groups was obviously higher than that in the normal control,but that in the treated group was remarkably lower than that in the infected group. Conclusion Apoptosis may be caused in testis spermatogenic cells with Toxoplasma infection,it may decrease after azithromycin therapy.
