摘要
以新疆杨 (PopulusalbaL var.pyramidalisBge.)叶片和腋芽为外植体 ,对适合植株再生的培养基、激素配比和培养条件进行了研究。结果表明 :对于叶片 ,添加 0 0 5mg LTDZ、0 0 3mg LIBA的 1 2MS培养基诱导出的愈伤组织容易分化成茎丛 ,愈伤组织转接到含 0 5mg LBA、0 0 5mg LNAA的 1 2MS培养基内产生大量的丛生芽。腋芽诱导茎丛的培养基为 1 2MS +BA1 0mg L +NAA1 0mg L。不定芽在IBA0 0 3mg L +1 2MS的培养基内可诱导产生大量的根 ,生根试管苗驯化、移栽后成活率可达 91%。对实验过程中的玻璃化问题进行了系统实验 ,发现大量元素中NH+ 4浓度过高和多次继代是引起玻璃化的主要原因 ,并提出了 3点防止措施。
The suitable medium, plant growth regulator and culture condition for regeneration of plants were investigated by using poplar leaves and buds as explants. For explants of leaf, 12 MS medium supplemented with 0.05 mgL TDZ and 0.03 mgL IBA could induce callus with high potential of organogenesis. When subcultured to 12 MS medium supplemented with BA and NAA (0.5 and 0.05 mgL respectively), this callus produced a great number of adventitious shoots. For explants of axillary bud, 12 MS medium supplemented with 0.1 mgL BA and 0.1 mgL NAA was the best for shoot proliferation. Rooting of shoots was obtained on 12 MS medium supplemented with 0.03 mgL IBA. Survival of seedlings after acclimatization was up to 91%. Vitrification during experiments was also investigated and methods to control the problem were recommended.
