摘要
为选取北虫草的最适液体培养基,以三因素三水平的正交试验设计培养北虫草的液体培养条件,北虫草菌种的接种量为5%(体积分数)。培养条件为18℃,pH为6.5,转速为120r/min,培养时间为72h。通过对培养所得的北虫草菌丝体干质量的测定分析可以得出培养北虫草的碳源最优水平为葡萄糖,氮源最优水平为NH4NO3,无机离子最优水平为K2HPO4。从而得到该菌种的最适液体培养基的配方(质量浓度)葡萄糖,NH4NO3,K2HPO4分别为20,1,1g/L。根据数据分析的R值和F值均可以确定:碳源为最重要的因素,应将其控制在最优水平,所以培养北虫草应该选取最优葡萄糖水平。实验结果为进一步大规模培养北虫草提供了重要的依据。
The optimum liquid medium on Cordyceps militaris was chosen. The orthogonal experiment was made with three factors and three levels of nutrient concentration to investigate the optimum liquid culture condition. The quantity of inoculation is 5%. The culture conditions are that temperature is 18 ℃,pH is 6.5,circle velocity is 120 r/min,culture time is 72 h.After the test and analysis of the desiccant weights of the mycelium, glucose was been chosen as the best level of carbon, NH_4NO_3 was been chosen as the best level of nitrogen, and K_2HPO_4 was been chosen as the best level of inorganic ion. The optimum liquid medium prescription is glucose 20 g/L,NH_4NO_3(1 g/L),K_2HPO_4(1 g/L).Form the analysis of the data, it can be sure that carbon is the most important factor. So it should choose the best level of glucose. The result of the study can supply important proof to culture cordyceps militaris in the larger level in the next.
