摘要
目的 观察氟对培养的大鼠破骨细胞基质金属蛋白酶 9(Matrixmetalloproteinase 9,MMP 9)的影响。方法 机械分离新生大鼠四肢长骨的破骨细胞 ,于分离后 2h加入不同浓度的氟化物 ,应用免疫组织化学技术和原位杂交技术检测氟化物干预下培养的破骨细胞MMP 9的蛋白质和mRNA的表达。结果 随着染氟剂量的增加 ,破骨细胞内MMP 9的蛋白质表达增高 ,以 2 0 ,4 0mg/LF 组最为显著 ,同时破骨细胞内MMP 9mRNA表达亦增加 ,1 0 ,2 0 ,4 0mg/LF 组与对照组比较差异有显著性。结论 氟通过提高MMP 9的蛋白质和mRNA表达 。
Objective To investigate the effects of fluori de s on matrix metalloproteinase-9 (MMP-9) in cultivated osteoclasts (OCs) in v it ro Methods OCs were isolated mechanically from long bones o f neonatal rats and cultured in 24-hole plate Two hours later, fluorides were a dded into the plate, the expressions of MMP-9 were investigated by immunhistoch e mistry and in situ hybridization techniques Results The e xpression of MMP-9 protein increased gradually with elevating fluoride concentr a tion, and more significantly in the 2 0, 4 0 mg/L fluoride groups than in co nt rol group In the mean time, an obvious rise of the expression of MMP-9 mRNA s ho wed up in the 1 0, 2 0, 4 0 mg/L fluoride groups, which groups differed sig nif icantly from that in the control group Conclusion The fluorid e spurs OCs and then enhances the bone resorption activities by increasing the MM P-9 expression of OCs not only at protein level but also at mRNA level
