摘要
目的 观察胶质瘤Wistar荷瘤大鼠γ刀照射后 ,C myc蛋白表达的变化 ,探讨γ刀的作用机理。方法 建立颅内种植胶质瘤C6细胞的Wistar荷瘤大鼠模型。 3周后 ,实验组给予γ刀治疗。计数荷瘤大鼠生存时间。免疫组织化学SP染色及计算机图像分析检测大鼠C myc基因蛋白水平表达。结果 实验组及对照组荷瘤大鼠生存时间分别为 :36 5± 3 1d ,2 8 0± 2 4d ;实验组生存时间明显延长 (P <0 0 1)。实验组及对照组荷瘤大鼠C myc基因蛋白阳性表达平均百分率分别为 :4 0 98%± 15 4 6 % ,77 6 2 %± 12 11% ;二者差异显著 (P <0 0 1)。结论 γ刀诱导胶质瘤凋亡的机制可能与抑制肿瘤细胞C myc基因蛋白水平表达有关。
Objective To observe the change of C-myc protein expression on glioma rats treated by gamma-knife. Methods Built Wister rat modle which bearing glioma C6 cells. Test group rats treated by gamma-knife after 3 weeks. Observe Wistar rats survival time. C-myc protein expression of Wistar rats were studied by immunohistochemical technique. Results Survival time of test group Wistar rats were 36.5±3.1 d, survival time of untreated group Wistar rats were 28.0±2.4 d. The survival time of test group was obviously prolonged(P<0.01). The C-myc protein expression rate in test group was 40.98%±15.46%, in untreared group was 77.62%±12.11%. The C-myc protein expression notably decreased in test group. Conclusion The mechism of gamma-knife induce glioma cells apoptosis maybe connectd with glioma cells C-myc protein expression decreased.
