摘要
目的 研究肿瘤坏死因子相关凋亡诱导配体 (TRAIL)受体在骨肉瘤细胞中的表达情况。方法 应用逆转录 聚合酶链反应 (RT PCR)对 2 2例骨肉瘤组织标本、MG 63骨肉瘤细胞株、U2 5 1脑胶质瘤细胞株以及正常人外周血淋巴细胞TRAILR1 R4mRNA表达进行检测。结果2 2例骨肉瘤组织标本中 15例同时表达TRAILR1、 R2和 R3 ,4例同时表达TRAIL R1和 R2 ,只表达TRAIL R1或 R2者 3例 ,所有标本中均未检测到TRAIL R4表达 ;MG 63骨肉瘤细胞株、U2 5 1脑胶质瘤细胞株以及正常人外周血淋巴细胞则均检测到TRAILR1、 R2和 R3的联合表达。结论 死亡受体TRAIL R1,R2在骨肉瘤中的普遍表达 ,是TRAIL诱导骨肉瘤细胞凋亡的分子基础 ;死亡受体和诱骗受体的差异性分布 ,并非TRAIL对选择性杀伤肿瘤细胞的关键性因素 ,可能还受其它因子调控。
Object To study the expression of tumor necrosis factor-related apoptosis-inducing ligand (TRAIL)-receptors (TRAILR) in human osteosarcoma.Methods The expression of TRAILR mRNA in 22 osteosarcoma tissues,peripheral blood leukocytes,osteosarcoma cell lines and glioma cell lines was detected by reverse transcription -polymerase chain reaction.Results Of 22 osteosarcoma tissues,15 were positive for TRAILR1-R3,and 4 were positive for TRAIL-R1 and TRAIL-R2.Of 22 osteosarcoma tissues,3 were only positive for TRAIL-R1 or TRAIL-R2,and 22 were negative for TRAIL-R4.Peripheral blood leukocytes,MG-63 osteosarcoma cell lines and U251 glioma cell lines were positive for all TRAILR1-R3.Conclusion The molecular basis of TRAIL inducing osteosarcoma apoptosis is the widespread expression of TRAILR1-R2 in human osteosarcoma tissues.The regulation of TRAIL pathway isn't according to the different distribution of death and decoy receptors of TRAIL but to the additional extra- or intracellular factors.
