外周血肝癌多药耐药荧光定量检测及意义 被引量:6

Significance of multidrug resistance in peripheral blood of patients with primary hepatocellular carcinoma detected by fluorescent-quantitative polymerase chain reaction

摘要 目的 评价荧光定量聚合酶链式反应 (FQ PCR)检测原发性肝癌外周血有核细胞多药耐药 (MDR1)基因表达与肝癌细胞MDR1基因表达的相关性 ,建立原发性肝癌外周血MDR1基因临床检测方法。方法 选择 5 5例原发性肝癌病例 ,术前抽血提取有核细胞 ,采用FQ PCR方法检测MDR1基因表达 ;采用逆转录 聚合酶链反应 (RT PCR)和流式细胞技术 (FCM)检测术后肿瘤标本MDR1mRNA和P 糖蛋白 (P gp)表达。结果  5 5例原发性肝癌病例荧光定量PCR、RT PCR、FCM方法检测MDR1的阳性表达例数分别为 2 5、3 0、3 3 ;3种方法的检测结果明显相关 (P <0 .0 1)。结论 FQ PCR方法检测外周血MDR1基因表达的结果与肝癌细胞MDR1的表达明显相关 ,可用于原发性肝癌病例多药耐药性的临床检测。 Objective To evaluate the significance of MDR1 mRNA in peripheral blood of patients with primary hepatocellular carcinoma by fluorescent-quantitative PCR.Methods Fifty-five cases of HCC were examined.The expression of MDR1 mRNA in peripheral blood was detected by FQ-PCR pre-operatively.The MDR1 mRNA and P-gp of resected tumor specimenwere detected with RT-PCR and FCM postoperatively.Results Of the 55 cases,25,30 and 33 were positive for MDR1 detected by FQ-PCR in peripheral blood,FCM and RT-PCR in resected tumor specimen,respectively.The expression of MDR1 detected in peripheral was significantly correlated with that in tumor specimen.Conclusion The expression level of MDR1 mRNA detected by FQ-PCR in pheripheral is coincidence with that in tumor specimen.The MDR1 mRNA in peripheral detected by FQ-PCR is suitable for clinical test to the patients with PHC.
