摘要
采用超临界二氧化碳流体萃取技术,提取了百合中的秋水仙碱,并用高效液相色谱法测定了萃取物中秋水仙碱含量;通过正交实验对萃取条件进行了优化筛选,确定了适宜的工艺参数.结果表明:百合粉经氨水碱化后,在40℃,18MPa,以乙醇作提携剂时萃取效果最佳.萃取物中秋水仙碱含量可由植物中的0.049%提高到6.40%.
The supercritical carbon dioxide fluid extraction of colchicine from the dry powder of lily was studied, and the extraction process conditions were optimized by the vertical-method. The contents of colchicine in the extracts were determined directly by the high performance liquid chromatography (HPLC). The result shows that the best process conditions are at 40 ℃, 18 MPa and with ethanol as pre-soaker, after lily powder is alkalified by ammonia liquor. The result also shows that, under the given conditions, colchicine concentration increases greatly from 0.049% in natural sample to 6.4% in the extracts obtained by the supercritical fluid extraction (SFE).
