摘要
所用的细胞按照Luben改良的方法培养.用S.D.大鼠颅盖骨,以胶原酶、胰蛋白酶消化,收集细胞,在10%胎牛血清的199培养液中,于5%CO2、37℃饱和湿度培养.在条件液中,分别加不同浓度的微量元素锌和钙,从形态学及生化指标探讨,证实微量元素锌和钙对成骨细胞样细胞的增殖分化都有明显的影响。
This study used bone cells were obtained from the calvaria of S. D. rats (3 week old) using essentally the methodology described by Luben with some minor modofications through isolation of collagenase I and four 20 mill treatment at 30 oscillations /mill at 37℃. The cells of the collection grew well at 0. 5mL cell suspension (2. 5 ×105 cell) in 24 multi well plates with 1mL 199 medium and 10% FBS. A total 2 ×10-6 bone cells/well was culture at 37℃ in 53% CO2+95% air. During the sixth day, the medium was then changed to conditioned medium containing Zn2+ . Ca2+. The experimantal contained 10-9 to 10-1. The result indicated that trace element stimulotes synthes is of the cell protein and stimulotes sensibly rise of the ALPase activity. they have a optimum peak volue of the trace elements concentration respectively and a optium administer phase of the trace elements.
