摘要
应用Ⅱ型胶原酶,37℃振荡60min(2次)消化分离胎鼠头盖骨,可获得25×10~5细胞。每瓶接种1×10~5细胞,经10％FBS-EMEM在37℃,5％CO_2培养箱中培养,获得生长理想的成骨细胞。细胞形态呈三角形、多角形和立方形。细胞膜碱性磷酸酶反应阳性。电镜X线分析显示含有一定量的钙。
In the present study 25 ×105 cells were obtained from the oalvaria of 22 days rat embryos through isolation of oollagenase II and two 60 mine treatment at 50 osoillations/min at 37℃. The cells grew well at 0.5ml cell suspension (1×105 cells)in culture flask with 3.5ml EMEM and 10% FBS. The osteoblasts were relatively large cells that exhibited basophilic cytoplasma and cytoplasmic processes. The processes joined with the processes of neighboring cells. The shape of osteoblasts appeared trigonal, polygonal and ouboidal. The membranes showed positive reaction with ALPase dye. The cells were demonstrated to contain, calcium by electric microscopic X analysis.
