成骨生长肽调节大鼠成骨细胞样细胞3种mRNA表达和电镜观察

摘要 研究了成骨生长肽(简称OGP)调节大鼠成骨细胞样细胞多种mRNAs表达和超微结构。将细胞分为OGP实验组和对照组,用RT-PCR方法检测细胞的I型胶原mRNA、骨钙素mRNA、碱性磷酸酶mRNA,并以GAPDH(磷酸甘油醛脱氢酶)作为内标,可求出各种mRNAs的相对含量。透射电镜观察二组细胞的超微结构。电镜观察到OGP组培养细胞的粗面内质网比对照组丰富,OGP组管状粗面内质网末端膨大成池,核糖体附在粗面内质网的表面上,细胞分化较完善。与对照组比较,OGP组分别上调细胞I型胶原mRNA、骨钙素mRNA、碱性磷酸酶mRNA表达。在10-12-10-8 mol/L OGP范围内,其浓度较高,细胞I型胶原mRNA和骨钙素mRNA表达也较高。结果表明OGP能促进细胞分化;上调细胞I型胶原mRNA、骨钙素mRNA、碱性磷酸酶mRNA表达。因此,0GP有促进骨形成功能。 To investigate the regulatory role of osteogenic growth peptide (OGP) on expression of Type I collagen, osteocalcin and alkalin phosphatase mRNAs and uhrastructural features in rat osteoblast-like cell. Cultured osteoblast-like cells were divided into experimental group treated with OGP and control group. Expression of Type I collagen mRNA,osteocalcin mRNA and alka-line phosphatase mRNA of cells were determined by RT-PCR method. Expression of mRNA of cells can be got by using glyceraldehyde-3-phosphate dehydrogenase (GAPDH) as the value of in-ternal reference. Uhrastructural changes of cells were observed under transmission electron mi-erescope. The Rough endoplasmic reticulum of cells were more abundent in the OGP group than that of in the control group. Swelling end of tubby rough endoplasmic reticulum turned to pool and ribosome on rough endoplasmic reticulum was observed in the OGP group. Differentiation of cells was excellent in the OGP group. Expression of mRANs of cells were higher in the OGP group and relatively lower in the control group. Expression of Type I collagen mRAN and osteocalcin mRNA of the cells in the OGP grouop were gradually upregulated with increasing concen-trations of OGP from 10-12 -10-8 mol/L. The results. revealed that the OGP can promote differ-entiation of cells and the Expression of Type I collagen mRAN, osteocalcin mRNA and alkaline phosphatase mRNA of the cells can be upregulated by exogenously administered OGP. There-fore, OGP can promote the function of bone formation in rat.
