摘要
对抗生素卡拉霉素和潮霉素进行菘蓝外植体抗性浓度筛选,结果表明不同外植体具有不同抗性.子叶芽分化阶段,卡拉霉素和潮霉素筛选浓度范围分别为10~50mg/L和5~30mg/L;下胚轴芽分化阶段的筛选浓度分别为15~50mg/L和10~30mg/L;分化苗生根时筛选浓度分别为30mg/L和20mg/L.芽分化阶段宜使用头孢霉素为抗菌剂,但分化苗生根时宜选用羧苄青霉素.
Through screening concentration of kanamycin and hygrimycin from culturing explants of Isatis indigotica Fort, the result showed that resistance concentration is different from different explant. In the stage of shoot regeneration from cotyledon, kanamycin is 10-50 mg/L, hygrimycin is 5-30 mg/L. In the stage of shoot regeneration from hypocotyls, kanamycin is 15-50 mg/L, hygrimycin is 10-30 mg/L. When plantlet grow in root medium, kanamycin is 30 mg/L, hygrimycin is 20 mg/L. In different stages of shoot or root regeneration, the best antibacterial agent is cefotaxime an or carbenicillin respectively.
