摘要
目的 :研究血管内皮细胞对骨髓间充质干细胞成骨能力的影响。方法 :体外分离培养大鼠骨髓间充质干细胞和肾血管内皮细胞 ,三维培养体系中直接、间接接触培养后 ,检测细胞中蛋白质和骨钙素含量及碱性磷酸酶活性。结果 :直接、间接接触培养及单独培养的骨髓间充质干细胞蛋白质含量为 (0 .195± 0 .0 2 3)mg/mL、(0 .174± 0 .0 15 )mg/mL、(0 .15 2± 0 .0 15 )mg/mL、(0 .14 1± 0 .0 14 )mg/mL ;碱性磷酸酶活性分别为 (0 .6 2 5± 0 .2 2 3) μ/mg、(0 .4 12± 0 .178) μ/mg、(0 .14 1± 0 .0 8) μ/mg ;骨钙素含量分别为 (0 .80 0± 0 .12 4 ) μg/L、(0 .4 35± 0 .2 16 ) μg/L、(0 .2 35± 0 .14 6 ) μg/L。经方差分析各组差异显著 (P <0 .0 5 )。 结论 :骨髓间充质干细胞与血管内皮细胞接触培养有良好的细胞相容性 。
Objective:To evaluate the effect of endothelial cell to marrow mesenchymal stem cell in in three-dimensional system.Methods:dissociation and culture rat bone marrow mesenchymal stem cell and endothelial cell,and in different co-culture models(co-culture with or without direct contact), Analysis ossification ability of bone marrow mesenchymal stem cell.Results:In directive and indirective co-culture and marrow mesenchymal stem cell alone culture ,Protein quantity is (0.239±0.023) mg/mL,(0.174±0.015) mg/mL,(0.115±0.012) mg/mL,(0.121±0.016) mg/mL; The ALP activity are (0.625±0.123) μ/mg、(0.412±0.178) μ/mg、(0.141±0.08) μ/mg;Osteocalcin are (0.8±0.124) μg/L、(0.462±0.149) μg/L、(0.235±0.146) μg/L.By statistics evaluation, To ossification ability of bone marrow mesenchymal stem cell co-culture is higher than culture alone(P<0.05),and direct is higher than indirect(P<0.05).Conclusions:Ossification ability of bone marrow mesenchymal stem cell can be regulated in co-culture with Endothelial cell by cytokine and cell membrane proteins.
