摘要
目的观察骨髓间充质干细胞(BMSCs)诱导的成骨细胞和肾血管内皮细胞直接共同培养的细胞相容性,并探索两者共同培养的合适比例。方法成骨细胞和血管内皮细胞按1:0、3:1、2:1、1:1、0:1比例分为5组直接共同培养,观察各组细胞形态并计数细胞比较增殖能力,检测碱性磷酸酶(ALP)活性,研究共同培养后成骨细胞成骨能力的改变。结果共同培养后细胞增殖及成骨细胞ALP染色阳性率高于单一细胞培养,成骨细胞与内皮细胞按2:1比例培养优于按3:1和1:1比例培养,增殖最快,ALP阳性率最高。结论成骨细胞和血管内皮细胞体外直接共同培养具有良好的相容性,可促进成骨细胞的增殖、分化及ALP的分泌,提高其成骨能力。两者按2:1混合是较佳比例。
Objective To study the influence of biological characteristics and optimal ratio of the osteoblasts induced from rabbits BMSCs and renal vascular endothelial cells direct co-culture in vitro. Methods Osteoblasts induced from BMSCs were co-cultured directly with renal vascular endothelial cells in different ratios of 1 : 0,3: 1,2: 1,1 : 1,0: 1. Osteogenesis was detected under an inverted microscope, cell count method, alkaline phosphatase activity assay respectively. Results The proliferation of the two kinds cells are all increased significantly than the single cell was cultured directly. The ALP activity of osteoblast was increased significantly than without culturing with endothelial cells. The proliferation and the ALP activity of the two kinds of cells in a ratio of 2:1 is higher than the others. Conclusion There were good cellular compatibility between the osteoblasts and endothelial ceils when the two kinds of ceils were co-cultured in vitro, moreover, enhance the proliferation and the function of the osteoblasts. Osteoblasts induced from rabbits BMSCs and renal vascular endothelial cells direct co-cultured in a ratio of 2:1 is optimal.
