乌司他丁和肿瘤坏死因子α抗体对大鼠肝脏缺血再灌注损伤的保护作用 被引量:11

Protective Effect of Ulinastatin and TNF-α Antibody on Ischemia and Reperfusion of Liver in Rats

摘要 目的 探讨乌司他丁和肿瘤坏死因子α(TNFα)抗体对大鼠肝脏缺血再灌注损伤的保护作用。方法 雄性SD大鼠 12 0只 ,分为正常对照组、缺血再灌注组 ,TNFα抗体处理组和乌司他丁 +TNFα抗体处理组 ,观察了肝脏缺血 60min及再灌注 1、3、6及 12h后 ,检测血清丙氨酸氨基转移酶 (ALT)、丙二醛 (MDA)及肝组织病理学变化。结果 肝脏缺血再灌注后 ,肝脏瘀血明显 ,血浆ALT和MDA含量均显著增高 ;肝脏缺血再灌注用乌司他丁 +TNFα抗体处理后 ,血浆ALT和MDA含量明显降低 ,肝组织瘀血减轻。结论 乌司他丁联合TNFα抗体可以抑制大鼠肝脏缺血再灌注所致的炎症反应 。 Objective To study the protective effects of ulinastatin and tumor necrosis factor-α(TNF-α) antibody on ischemia and reperfusion injury of liver in rats. Methods One hundred and twenty male SD rats were randomly divided into four groups: the normal control group, ischemia and reperfusion group, TNF-α antibody group and ulinastatin plus TNF-α antibody group. And the animals were killed after 60 minutes ischemia of liver followed by reperfusion for 1,3,6 and 12 hours. Serum alanine aminotransferase(ALT) and malondialdehyde(MDA) were detected, and liver histopathologic lesions were observed. Results After ischemia and reperfusion, the serum level of ALT and MDA remarkedly increased, and the hepatic congestion was prominent. Treatment of ulinastatin and TNF-α antibody could decrease the serum level of ALT and MDA significantly, and relieve hepatic congestion. Conclusions Ulinastatin and TNF-α antibody can suppress the inflammatory reaction induced by hepatic ischemia and reperfusion, and has protective effects on rat hepatic ischemia and reperfusion injury.
