摘要
目的 :探讨叶绿素衍生物CPD4光动力学治疗杀伤体外培养卵巢癌细胞的作用机制。方法 :应用不同浓度CPD4、不同能量激光对人卵巢癌细胞株 3AO进行光动力学治疗 ,在光镜和电镜下观察其形态变化 ;做DNA片段分析 ;并用流式细胞仪分析细胞DNA含量的改变和细胞凋亡率。结果 :CPD4光动力学治疗明显抑制体外培养人卵巢癌细胞株 3AO的生长 ,细胞分裂阻滞于G0 ~G1期 ,并诱导细胞发生凋亡。凋亡细胞表现为细胞固缩 ,染色质聚集或断裂 ;DNA凝胶电泳显示梯形条带。结论 :凋亡为叶绿素衍生物CPD4光动力学治疗杀伤体外培养细胞的主要作用机制。
Objective:To investigate the mechanism of chlorophyll derivative CPD4 photodynamic therapy of cultured ovarian cancer cells.Methods:The human ovarian cancer cells 3AO were treated by photodynamic therapy with different CPD4 concentration and different laser energy.Morphologic changes were examined by light and electron microscopes.DNA fragment were analyzed.Changes of nuclear DNA contents and percent of cell apoptosis were detected by flow cytometry.Results:The growth of human ovarian cancer cells 3AO was inhibited after CPD4 photodynamic therapy.Cell cycle arrested at G 0 G 1 phase and apoptosis was induced.The morphology of 3AO showed nuclear chromatine condensation and fragmentation.Typical ladder pattern of DNA fragmentation was observed.Conclusion:Apoptosis is the main mechanism of chlorophyll derivative CPD4 photodynamic therapy to kill culture cells in vitro.
